Purpose: Previous studies demonstrated that curcumin has widely anti-inflammatory effect and protects vascular endothelial cell against injury. To observe the effects of curcuma decoction on heat shock protein 27 (HSP27) and tumor necrosis factorα(TNF-α) in rats with vascular endothelial injury, and to explore the molecular mechanism of protection effects from the aspect of inflammatory reaction,for PCI after"restenosis"to provide experimental evidence for prevention and treatment.Material and method:Analog PCI operation to cause acute vascular intimal injury, in which turmeric group 36 and saline group 36 and 36 with the sham group and 36 normal group as control. Turmeric sets of the general routine PCI after turmeric (100mg/Kg/d) administered orally; saline treated group after the general conventional analog PCI saline 3ml / d orally; sham laparotomy suture does not insert the balloon catheter; normal control group did not use any intervention. PCI simulation at 3,7,14 days after the first use of light and electron microscopy changes in the structure of rat endothelial cells; detected by ELISA in 144 serum and the hot shock protein 27 and tumor necrosis factor alpha.Results:1. Light microscope, turmeric group 7 days after injury medication 7 days compared with model group, endothelial loss of local vascular injury and intimal damage slightly reduced, the degree of vascular stenosis can be seen to reduce. And normal control group, but there are still differences in lumen area were significantly smaller than the undamaged side.2. Electron microscope can be seen in the third day after balloon injury irregular columnar endothelial cells, cell shrinkage, cytoplasmic stain, nuclear fragmentation. Shows that the release of a small number of apoptotic bodies. Seventh day, cell necrosis increased significantly. Model group obviously necrotic cells were observed, there are a lot of fat soluble lipid and necrotic material, and turmeric fried group cell necrosis and the other control group significantly decreased apoptosis is only a little debris.3. Analog PCI rats after acute vascular intimal injury caused by acquired 3d,7d,14d after the Fried turmeric group TNF-αserum levlels were significantly lower than model and sham operation group (P < 0.05),the 14d group to the most significant(P < 0.05).HSP-27 of the Turmeric group in 7-14d was significantly higher than serum levels of the model and operation group (P < 0.05).4 Serum levels with HSP-27 and TNF-αexpression was negatively correlated.Conclusion:1. Using light and electron microscopy after simulation of PCI changes in the structure of endothelial cells in rats shows that turmeric endothelial protective effect of a relatively clear.2. Chinese medicine turmeric intervention of the rats was significantly decreased serum levels of the TNF-α,reduce the damage to the vascular endothelial cell inflammatory response, while making as a protective agent in vascular endothelial cells in endothelial cell damage HSP-27 to achieve significantly increased peak, rapid recovery of endothelial function, to maintain maximum structural integrity of vascular endothelial.3. Apoptotic factors are involved in the pathology of restenosis after PCI physiological processes. Regulation to TNF-αof the HSP-27 may be involved in the repair of the endothelial cells.
