Objective To study the inhibition and the mechanism of common turmeric (wenyujin) on human gastric carcinoma cell line SGC-7901 in vitro.Methods The common turmeric oil was obtained by supercritical fluid extraction (SPE). With the 5-FU as the positive control, the effects of the extract of common turmeric on proliferation of human gastric carcinoma cell line SGC-7901 in vitro by using MTT assay were studied. The levels of insulin-like growth factor I (IGF- I ) and insulin-like growth factor II (IGF- II ) in supernatant were detected by radioimmunoassay. The effects of common turmeric on vascular endothelial growth factor (VEGF) mRNA expression inhuman gastric carcinoma cell line SGC-7901 were investigated by employing the semi-quantitative method of reverse transcription polymerase chain reaction (RT-PCR).Results By supercritical fluid extraction we get the extract 1 and extract 2 of common turmeric. Compared with the negative control, extract 1 and extract 2 of common turmeric can significantly inhibit the growth of human gastric carcinoma cell line SGC-7901 in vitro with certain concentration(all P < 0.01). Extract of common turmeric of 25,50,100,200ug/ml exerted the inhibitory actions on SGC-7901 cells, especially at 200ug/ml. In 200ug/ml level, the inhibitory rate was 64.44% and 60.80%, respectively, there were no different between the extract of common turmeric and 5-FU in their inhibition on SGC-7901 (P > 0.05). Extract 1 and extract 2 of common turmeric can significantly decrease the expression of IGF-1 and IGF- II in supernatant compared with negative controls(all P < 0.01). Human gastric carcinoma cell line SGC-7901 analyzed expressedVEGF121 and VEGF165, incubated with different Concentration of extract of common turmeric, the expression VEGF121 mRNA and VEGF165 mRNA in SGC-7901 cell deceased. At the concentration of 100ug/ml, the expression of VEGF121 mRNA and VEGF165 mRNA were decreased gradually(all P < 0.01).Conclusion We conclude that common turmeric could inhibit the growth of human gastric carcinoma cell line SGC-7901 in vitro, the decrease of IGF- I , IGF- II and VEGF expression might be the primary mechanism.
