| Objective: To investigate the Toll-like receptor-mediated NF-κB signaling pathway effect of glycyrrhizin for Ulcerative Colitis.Methods: Selected 49 healthy SD rats, the 10 rats were as the normal control group(group A), and the remaining 39 rats were combined modeling TNBS / ethanol prepare legal ulcerative colitis and then were randomly equally divided into model group(group B), sulfasalazine group(group C), glycyrrhizin group(group D) that were given saline, sulfasalazine and glycyrrhizin intervention treatment.Results: 1.The naked and pathology observed the ulcerative colitis in rats with colorectal cancer model was successful. Were observed before and after rat model and intervention treatment after the hair, feces traits, bloody and weight is a bigger change. But contrast intervention treatment of two groups of willow nitrogen sulfonyl pyridine group(group C) and compound glycyrrhizin glucoside group(group D) rats between hair, feces traits, bloody and weight change.2.The serum expression of TLR4 and p65 in the glycyrrhizin group were compared with the control group and the sulfasalazine group were showed no significant difference,significantly lower than the model group(P <0.05)。3.The P65 and TLR4 protein expression of the sulfasalazine group were the compared difference among the glycyrrhizin group and the normal control group were not statistically,significant significantly lower than the normal model group(P <0.05)。4.Pearson correlation analysis showed that p65 expression and the TLR4 expression in ulcerative colitis model showed the positively correlation(P <0.05).Conclusion: Glycosides compound glycyrrhizin in the treatment group rats serum level of TLR and nf-kappa B p65 contrast to model control group decreased, compared with no difference between the willow nitrogen sulfonyl pyridine group; Colon tissue protein positive rate is significantly lower than the model control group, compare the willow nitrogen sulfonyl pyridine group, no significant difference shows that compound glycyrrhizin glucoside blocking toll-like receptor mediated the nf-kappa B signaling pathway.Toll-like receptor-mediated signaling pathways of NF-κB plays an important role in the development of inflammatory bowel disease, glycyrrhizin can blockedthe expression of the TLR pathway and p6, thus provides a theoretical basis for clinical use. |
