Objective: by separating culture of intestinal mucosal epithelial cells, given different composition ratio of the gut contents and specific TLR antibody, detection comparison of intestinal mucosa epithelial cells of TLR4 and NF kappa B expression and cytokine changes, visually verify effect of different intestinal content and TLR antibodies of TLR mediated immune responses. Methods: the body mass is about 300 g in 20 healthy rats, were randomly divided into normal group(n=10) and UC group(n=10), on the fourth day after operation, was cultured from the colon segment, with different proportion of bacteria or intervention treatment of intestinal epithelial cells, respectively different interventions are divided into corresponding groups: group A: control group(no intervention); group B: normal group(normal intestinal cavity bacteria added intestinal cavity mucosa flushing fluid); group C: normal intestinal bacteria membrane group(adding normal intestinal mucosal suspension); group D: UC model intestinal flora group(UC animal model of intestinal mucosa lavage); E group: UC model group(intestinal bacteria into intestinal mucosa UC animal model suspension); group F: the anti TLR antibody group(with anti TLR antibody). Then the cells were collected to detect the expression of NF- and TLR4 nuclear factor kappa B, and the supernatant was collected to detect the level of cytokine IFN- and IL-1.Result, reduce the level of inflammatory factor and inflammatory factor, inflammatory factor group increased significantly compared with the results: D, e group and a group, TLR4 and NF kappa B m RNA and protein levels and IFN- and IL-1 beta inflammatory factor level, D, e and B, C group compared to the TLR4 and NF kappa B m RNA and protein levels and IFN- and IL-1 levels were significantly elevated in group A and B, C group compared with no statistical significance in group F and D group of TLR4 and NF kappa B protein levels decreased significantly, NF kappa B m RNA and TLR4 protein levels, IFN gamma and IL-1 levelswere not significantly changed in Group F and E group TLR4 m RNA and TLR4 and NF kappa B protein levels of IFN gamma, NF kappa bmrna IL-1 levels had no significant change. Conclusion: Genesis of abnormal immune reaction in the ulcerative colitis and intestinal contents of stimulation and TLR4 monoclonal antibody can down-regulation of TLR4 mediated immune pathway and its downstream inflammatory factor, can suppress the occurrence of immune response.
