| Objective Establishing spleen-kidney yang deficiency UC animal models,the combination of disease and syndrome,and to discuss the essence of TCM spleen-kidney yang deficiency from molecular levels,hoping to find its modern scientific foundation.Sishen Pill is the classical prescription of warming the kidney and strengthen the spleen.According to the curative observation of Sishen Pill,and its effect on the gene expressions of positive and negative regulatory factors on the TLR/IL-1 signaling pathway.Parts of the mechanism of treating UC by warming the kidney and strengthen the spleen will be further revealed.Also,more experimental evidence will be offer for TCM in UC’s treatments.Methods To made UC models by using the method of gavaging with Fanxieye,intraperitoneal injection with hydrocortisone and clysis with TNBS/ethyl alcohol.The behavior and damage in colon tissue were observed with naked eye,pathological sections were examined and to do an assessment for animal model and Sishen Pill’s efficacy.TLR4,IRAK-M and Tollip are positive and negative regulatory factors on TLR/IL-1signal pathway.The relative mRNA expressions of the factors in UC colon tissue were detected by RT-qPCR method.Results ①The assessment of UC models:The symptomatic score in model group was higher than blank group obviously(p<0.01),also the score of colon tissue(p<0.01);Colonic mucosa with inflammatory cells infiltration under light microscope.The expressions of T,FT3,FT4,IL-2 in model group’s serum were lower then the blank group,but the IgE was higher(p<0.01);SOD decreased while MDA increase(p<0.01).②The assessment of Sishen Pill’s efficacy:Sishen Pill can improve the symptoms and signs of UC,The sign scores in Sishen group are lower than those in model group(p<0.01),also the colon tissue observed by naked eye.Inflammatory cells were infiltration on Colonic mucosa observed with light microscope.Compared with the model group,the expressions of T,FT3,FT4,IL-2 in model group’s serum increased but the IgE decreased(p<0.01);SOD higher while MDA lowe(p<0.05).③TLR4,IRAK-M,Tollip mRNA:Compared with the blank group,the positive expressions of TLR4,Tollip mRNA in rats’ colon tissue in model group were at higherlevels.After the medical threatment,the expressions got down in Sishen group(p<0.01).On the contrary,the expression of IRAK-M mRNA was opposite(p<0.01).Conclusion It is successful to make UC models by using the method of gavage with Fanxieye,intraperitoneal injection with hydrocortisone and clysis with TNBS/ethyl alcohol.The medical basis of spleen-kidney yang deficiency relates to immune imbalance,hormone disoder,functional impairment in hypothalamus-pituitary-adrenal,and so on.Sishen Pill,warming the kidney and strengthen the spleen,treats UC well,especially in the chonic stage.Some of the mechanisms are to adjust TLR4,IRAK-M,Tollip mRNA.In order to regulate the balance in yin-yang and to restore the steady state in immune system,warming the kidney and strengthen the spleen can treat UC by means of adjusting the activity of regulatory factors in TLR/IL-1 signaling pathway. |
