ObjectiveTo explore the differentiation of bone marrow mesenchymal stem cells into the protective effect and mechanism of local transplantation of neuron like cells on spinal cord injury in ratsof safflower yellow in vitro, for the clinical treatment of spinal cord injury(SCI) and provide a theoretical basis.Methods50 male SD rats were randomly divided into normal group, model group, safflower yellow group, MSCs group, treatment group. Constructed the SCI rat model by improved Allen method. The normal group was treated with routine feeding, no treatment; model group in phosphate buffer solution damage local injection of saline and equal; safflower yellow group in phospha-te buffer solution damage local injection of Safflor and equal; the group MSCs, partial damage MSCs transplantation; in the treatment group were given local injury in neuron like cells in vitro induced by transplantation, once everyday. Using the modified BBB score examination on animal behavior, measurement of grip, posture maintaining capability, the recovery of spinal cord function evaluation of injury, immunofluorescence detection of GFP-GFAP, GFP-Neu N and NGF, MDA, SOD,test the cell apoptosis by TUNEL.Result(1) The BBB score of each group compared with the normal group in section 24 h, 7d, 14 d, 28 d were significantly decreased, the difference was statistically significant(P<0.01); each group compared with the model group in same period, the difference was statisticallysignificant(P<0.05);each group compared with safflower yellow group in the same period of, the difference was statistically significant(P<0.05); each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).(2) The number of GFAP positive cells in each group compared with the normal group in the 24 h,7d, 14 d, 28 d were increased obviously, the difference was statistically significant(P<0.01);except 24 h, each group compared with model group in the same period, the difference was statistically significant(P<0.05); except 24 h, each group compared with safflower yellow group in the same period, there was statistically significant differences(P<0.05);except 24 h, each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).(3)The number of NGF positive cells in each group compared with the normal group in the 24 h,7d, 14 d, 28 d were increased obviously, the difference was statistically significant(P<0.01); each group compared with model group in the same period, the difference was statistically significant(P<0.05);each group compared with safflower yellow group in the same period, there was statistically significant differences(P<0.05);each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).(4)The number of Neu N positive cells in each group compared with the normal group in the 24 h,7d, 14 d, 28 d were increased obviously, the difference was statistically significant(P<0.01); each group compared with model group in the same period, the difference was statistically significant(P<0.05);each group compared with safflower yellow group in the same period, there was statistically significant differences(P<0.05);each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).(5)The level of SOD of each group compared with the normal group in section 24 h, 7d, 14 d, 28 d were significantly decreased, the difference was statistically significant(P<0.01); each group compared with model group in the same period, the difference was statistically significant(P<0.05);each group compared with safflower yellow group in the same period, there was statistically significant differences(P<0.05);each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).(6)The level of MDA of each group compared with the normal group in section 24 h, 7d, 14 d, 28 d were significantly decreased, the difference was statistically significant(P<0.01); each group compared with model group in the same period, the difference was statistically significant(P<0.05);each group compared with safflower yellow group in the same period, there was statistically significant differences(P<0.05);each group compared with MSCs group in the same period, the difference was statistically significant(P<0.05).Conclusion(1) Bone marrow mesenchymal stem cells can be divided into neuron like cells in vitroinduced by safflower yellow.(2) Neuron like cell transplantation can improve BBB score of rat with spinal cord injury model.(3) The pathological changes of neuron like cell transplantation can relieve spinal cord injury model in rats, the increased expression of GFAP, Neu N, NGF, reduce glial scar formation,promote the recovery of spinal cord neuronal regeneration and functional damage.(4)Though increaseing the content of SOD, decreaseing the content of MDA may be a possible mechanism of neuron like cell transplantation for treatment of spinal cord injury.
