Objective:Along with the diversification of the range of human activities and their way of life, more and more accidents happened, deaths because of excessive blood loss has attracted more and more attention. How we can avoid such things from happening after the injury occurred, highly effective hemostatic materials in a timely manner is essential. Hemostatic products now on the market mainly consist of oxidized regenerated cellulose like surgical, collagen protein, chitosan and alginate salts, zeolite. Different materials have different hemostatic mechanisms, mostly work slow and do not meet the requirement of emergency. Chitosan have hemostatic, antimicrobial and healing biological functions, it is a degradability material, has been widely used in biomedical applications. This experiment based on chitosan, prepared chitosan powder through chemistry modification, finding its physical-chemical properties and liquid absorption properties. Assessing its biological safety and effectiveness to stop bleeding, inorder to achieve the goal of rapid blood condensation, and rapid hemostatic effect.Method:1. Study the physica-chemical properties of chitosan powder, including the degree of deacetylation and carboxylation, ash content, heavy metal content, water content and infrared spectrum; 2. Testing its speed and absorption capacity to liquids compared with Celox powder, evaluation the absorptive effects. Established femoral artery model in New Zealand rabbits, recording the hemostasis time, observe the effectiveness of rapid hemostasis and evaluate the hemostatic effect compared with Celox powder; 3. Evaluate the security of the biological material through the skin irritation test and cytotoxicity test.Results:1. The physical-chemical properties of chitosan hemostatic powder meet the industry standards; 2. The absorption of chitosan powder has been noticeably improved, absorption of ion water, absorption of normal saline and anti coagulation artery blood are obviously good than Celox powder. Compared to Celox powder, the effect of chitosan hemostatic powder is better, less time to hemostasis and no repeat bleeding; 3. The new powder meets the requirements of intracutaneous irritation, no cell toxicity, and its biosecurity meet the standard of biomedical materials.Conclusion:Compared with Celox powder, chitosan powder has quicker absorption rate, more liquid absorption(particularly the absorption of arterial blood), combining bio-safety and hemostasis. It’s a potential biological material.
