Objectives To observe the toxicity effect of Euphorbia kansui with Licorice on the rats’ kidney, and the relationship with the proportion of Licorice. For suppling reference information for clinical safe medication.Methods To separate 50 8-week SD rats into five groups including normal group, drug groups with different proportion of Euphorbia kansui and Licorice 1:0,1:0.25,1:0.5,1:1, with 10 mice in each group. the dose of Euphorbia kansui was 2g/kg/d in each group,(concentration of Euphorbia kansui:0.4mg/kg/d). The experiment groups were given the corresponding drugs and lasted for 28 days. The volume were 0.5ml/kg/d, the normal group were gived to the same volume of distilled water.and detect the general conditions of the the rats.These rats were all executed 24 hours after the last dose, and their bilateral kidneys were taken to weight the kidney observe,and calculate the index of kidney. The morphological changes in kidney were observed by HE staining. SCr and BUN of blood were measured by omni-biochemistry device. The expression of Bcl-2 and Bax proteins in kidneys were detected by immunohistochemistry technique.The total kidney m RNA was extracted by RT-PCR technique for measurement of the expression of Bcl-2 and Bax m RNAs.Results 1 The rats general condition: Except the normal group,the other group rats were all in low spirits,shortness of breath,squint,to walk unsteadily and so on after given the corresponding drugs.The most obviously group was 1:0 group, the toxicity effect had negative correlation with the doses of Licorice.2 The effect on the changes of visccra index of the kidney: Compared with the normal group,the expression of kidney index were significantly increased in the other groups(P<0.05 or P<0.01). the 1:0 group were higher than 1:0.25 group,1:0.5 group,1:1 group(P<0.05 or P<0.01).Followed in the level of:1:0 group>1:0.25 group>1:0.5 group>1:1 group.3 The changes of rats on the SCr and BUN content: Compared with the normal group,the content of SCr and BUN were significantly increased in the other groups(P<0.05 or P<0.01). The 1:0 group were higher than 1:0.25 group,1:0.5 group.1:1 group(P<0.05 or P<0.01).Followed in the level of:1:0 group>1:0.25 group>1:05 group>1:1 group.4 The influence on kidney tissue structure:Rats cortex and medulla were clear boundaries in each groups.The glomerularstructure is clear,and not seen obvious atrophy and hyperplasia.The capillary is not seen expand and hyperemic,and the afferent arteriole no engorgement and hyaline degeneration, The collecting duct epithelial cells no degeneration and necrosis, edema.Compared with the normal group,there were not seen obvious change in rats glomerular and collecting duct. Compared with the normal group,the other groups rats proximal tubule and distal convoluted tubule were significantly occur edema.Their epithelial cells were increase in size,the gap is not clear,The cytoplasm is filled with Pale pink particles,cell lumens stenosis and irregular.The serious of the 1:0 group was obviously.5 The effect on the changes of Bcl-2 of Bax proteins and m RNAs expression on rat’s kidney: Compared with the normal group.the expression of Bcl-2 protein and m RNA were significantly decreased in the other groups(P<0.05 or P<0.01). the 1:0.25 group,1:0.5 group,1:1 group were higher than 1:0 group(P<0.05 or P<0.01).Followed in the level of:1:1 group>1:0.5 group>1:0.25 group>1:0 group. Compared with the normal group.the expression of Bax protein and m RNA were significantly increased in the other groups(P<0.05 or P<0.01). the 1:0.25 group.1:0.5 group,1:1 group were lower than 1:0 group(P<0.05 or P<0.01).Followed in the level of:1:0 group>1:0.25 group>1:0.5 group>1:1 group.Conclusions Euphorbia kansui had obvious toxicity on the rats’ kidney.The Licorice could restrain the Euphorbia kansui’s toxicity to rats’ kidney.And in significant dose dependent manner.Whose restrain function had a positive correlation with the doses of Licorice.
