This study determined the contents of daidzein, glycitein and genistein in Edamame, fermented Edamame and soybeans. Soybean isoflavones in the samples were hydrolyzed by acid reflux, and daidzein, glycitein and genistein were quantified by high-performance liquid chromatography(HPLC).This analysis was carried out on a Hypersil ODS2(250mm×4.6mm, 5μm) column, methanol- 0.3% phosphoric acid solution volume fraction(V(methanol) :V(mass fraction of 0.3% phosphoric acid) = 48:52) as the mobile phase, column temperature at 25 ℃, the detection wavelength was 260 nm, the flow rate was 0.7mL/min. The content of daidzein in Edamame, fermented Edamame and soybeans was 255.96, 286.04 and 358.08μg/g; the content of glycitein was 57.96, 65.51 and 59.73μg/g; and the content of genistein was 12.88, 26.40 and 4.62μg/g. The content of genistein in fermented Edamame increased considerably, it is 2.05 and 5.71 times in Edamame and soybeans respectively. After fermentation, the three kinds of soybean isoflavone content have increased, but the increase of genistein was significantly.The total polyphenols in Edamame, fermented Edamame and soybean were determined by Folin-Ciocalteu colorimetric. As a result, The total content of polyphenolic compounds in fermented Edamame((3.31±0.138)mg/g) is about 2.9 times than that of soybeans((1.148±0.028)mg/g), and is about about 1.2 times than that in Edamame((2.69±0.011)mg/g).Through the determination the contents of soy isoflavone aglycone in Edamame, fermented Edamame and soybean and the contents of the total polyphenols in three samples, we found that the antioxidant activity of fermented Edamame is higher than that of Edamame and soybean. In order to test our inference is correct, the antioxidant activities of ethanol extracts from Edamame, fermented Edamame and soybeans in scavenging of DPPH and ABTS radicals and reducing power were assessed as well. In terms of Anti-oxidative activity, the ethanol extract from fermented Edamame had stronger antioxidant activity in scavenging of DPPH and ABTS radicals than that from Edamame and soybeans. The order of reducing power is fermented Edamame > Edamame > soybeans. Therefore, fermented soybean had a strong antioxidant activity.This paper studied the effect of fermented soybean milk on streptozotocin(STZ) induced type 2 diabetic mice. Type 2 diabetes mellitus was induced in mice by intraperitoneal injection of streptozotocin(STZ, 100mg·kg-1). Mice were randomly divided into control group, model group, positive control group(metformin hydrochloride, 250 mg ? kg-1), fermented Edamame milk low dose(1.5g ? kg-1), fermented Edamame milk high dose(2.25 g ? kg-1).All mice were intragastic administration for 4 weeks, body weight was measured every two days, food and fluid intake were monitored every day. Finally, the fasting blood glucose and oral glucose tolerance of mice were measured. The mice were sacrificed to collect the blood to determine the serum lipids, The mice were dissected and taken the liver, spleen, kidneys, thymus and pancreas. Organs were weighed and calculated organ index. Simultaneous determined the SOD activity and MDA content in liver tissue. The results showed that low doses of fermented Edamame milk can significantly reduce fasting blood glucose in mice(P <0.01) and increase the area under the glucose AUC line oral glucose tolerance(P <0.01); Low dose of fermented Edamame milk can significantly reduce type 2 diabetes in mice serum TC, TG levels(P <0.01); while significantly improving liver tissue SOD activity(P <0.01), reducing MDA content(P <0.01); low dose also can be a good repair the damage the organs of mice. High doses of fermented Edamame milk can improve the impact of type 2 diabetes in certain extent, but overall, the low dose of fermented soybean milk is better.This study is to determine the effects of fermented edamame milk on the antioxidant capacity in aging mice brain and skin hydroxyproline content. Through the back of the neck subcutaneous injection of D- galactose to establish subacute aging model mice, fermented edamame beany milk in a dose of 1.25 g·kg-1, 5 g·kg-1orally aging mice.56 days later, to determine the brain tissue total antioxidant capacity(T-AOC), malondialdehyde(MDA) content, nitric oxide(NO) content and the skin hydroxyproline(HYP) content of aging mous. Low dose of fermented edamame milk can significantly increase the aging mouse brain tissue total antioxidant capacity(P <0.05), both doses can reduce brain tissue MDA content(P <0.01), High dose has a significant effect in reducing the nitric oxide content(P < 0.01).Each dose of Fermented edamame milk could significantly increase in aging mice hydroxyproline content(P <0.01), the effect of low dose is better. Fermented edamame milk can increase the antioxidant capacity in aging mice brain tissue and have a protective effect to aging mice skin tissue.
