Diabetes are inflammatory related diseases.Based on the researches on the anti-oxidant,anti-inflammatory and anti-insulin resistance effects of Pueraria isoflavone,the anti-effects of the less researched compound of them,formononetin,on type ? diabetes mellitus was studied.These studies could lay the pharmaceutical foundation for the selection of anti-diabetes drugs from Pueraria isoflavones.In the anti-oxidant researches,the direct cleaning effects of DPPH free radicals,hydroxyl radical,peroxide and superoxygen anion radicals of 11 kinds of Pueraria isoflavone compounds,puerarin,daidzein,3'-methoxy puerarin,genistein,tectorigenin,formononetin,ononin,biochanin A,genistin,puerarin apigenin and glycitein,were investigated to compare the antioxidant abilities of these isoflavones.And it was found that all of the 11 kinds of Pueraria isoflavone compounds express certain direct free radical cleaning effects,of which the effects of genistein and tectorigenin are strongest.In the anti-insulin resistance researches,the anti-insulin resistance effect of 6kinds of Pueraria isoflavones,puerarin,genistein,tectorigenin,formononetin,biochanin A,and glycitein,on the insulin resistance of HePG2 was detected by Glucose kit(glucose oxidase method)to measure the glucose consumption of the medium.And it was found that all of the 6 Pueraria isoflavones express certain anti-insulin resistance effect for hepatocytes.In the anti-oxidant researches,the anti-NO release effect of 6 kinds of Pueraria isoflavones,puerarin,genistein,tectorigenin,formononetin,biochanin A,and glycitein on LPS-activated macrophages was detected by Griess method.And the influence of 3 kinds of Pueraria isoflavones,tectorigenin,biochanin A,and formononetin,on two inflammatory cytokines(IL-1? and TNF-?)in cell culture supernatant fluids of LPS-activated macrophages was investigated by Griess method.It was found that all of the 6 kinds of Pueraria isoflavones express some degrees of inhibiting effects on the activation of macrophages on rather high concentration and all of the 3 kinds of Pueraria isoflavones express some degrees of reducing effects on the production of cytoinflammatory factors,TNF-?and IL-1?.Only the effects were rather weak.In the anti-type ? diabetes mellitus researches,the model mice of type ? diabetesmellitus was established by high sugar and high fat feeding and STZ injection,and one of the Pueraria isoflavones,formononetin,was used as the research object and glibenclamide was used as positive drug.After the model mice had been successfully established,they were divided into parallel groups of diabetic control group,positive drug group,low dose group,middle dose group and high dose group,and another 10 mice were used as normal control group.Formononetin and positive drug were continuously intraperitoneally injected for 4 weeks.After the injection,three physiological indexes(the average body weight,dietary intake and drinking water of each group),and five biochemistry indexes(fasting blood glucose,triglyceride,total cholesterol,low,high density lipoprotein cholesterol)were recorded every week.The serum creatinine was also detected to analyse the effects of formononetin on the improvement of renal function.Data were analyzed statistically by SPSS 20.0.And it was found that formononetin could improve glucose metabolism disorders and lipid metabolism,reduce the levels of blood glucose,triglycerides,total cholesterol,low-density lipoprotein cholesterol of model mice and increase high-density lipoprotein cholesterol levels.Formononetin could also alleviate the symptom of "three indexs were high while one index was low" in diabetic mice.Formononetin also had certain protection effects on renal function which could delay the elevation of serum creatinine content in type ? diabetes mellitus mice.Conclusion:Pueraria isoflavones process certain anti-oxidant,anti-inflammatory and anti-insulin resistance effect.Formononetin,one of the Pueraria flavonoid compounds,has some functions of alleviating the sugar metabolism disorder and fat metabolism disorder in type ? diabetes mellitus mice,and improving Renal function,so reducing the symptoms of type ? diabetes.
