Vernonia esculenta Hemsl. belongs to the Compositae, shrub or small tree, distributed in many parts of China and South Africa, the pith of this plant can be eaten, and it’s leaves can cure hot burns. In this paper, we make more systematic study for the fruit from Vernonia esculenta Hemsl., and which focus on the chemical composition, extraction of the process, the role and mechanism of hypoglycemic activity for the fuit from Vernonia esculenta Hemsl.. Provide reference for the development and utilization of resources in Vernonia esculenta Hemsl..In this study, we use the percolation extraction method to separate for the chemical composition of the fruits from Vernonia esculenta Hemsl..We get Petroleum ether extracts, ethyl acetate extracts, 95% ethanol extracts, 70% ethanol extracts and 30% ethanol extracts, The residue was extracted by ultrasonic extraction method to obtain an aqueous extracts, then the ethyl acetate extracts were separated by column chromatography, thin layer chromatography and high performance liquid chromatography, and so on, which obtained 11 chemical compounds, and 8 compounds were identified finaly, it’s Vernolic acid, Vernodalin, Ursolic acid, Vernodol, Luteolin, Butin, β-sitosterol and Protocatechuic acid, which were isolated from this plant, which have greater content of Vernolic acid, Vernodalin and Vernodol.In this study, we use UV spectrophotometry to measure the flavonoid content of Vernonia esculenta Hemsl., results show that: The optimum extraction conditions were 70% ethanol, solid/liquid ratio 1:20, extraction time 60 min / times, extracted four times. Under this condition, the average of the total flavonoids yield is higher, which were higher than the orthogonal experiment.In this study, spontaneous mouse model of type 2 diabetes was used as the research object, the fruit extract from Vernonia esculenta Hemsl.. Were seperited into petroleum ether, ethyl acetate, 95% ethanol, 70% ethanol and 30% ethanol group, By detecting mice fasting blood glucose, random blood glucose, weight, insulin resistance and impaired glucose tolerance and other changes, to investigate the hypoglycemic activity of the fruit from Vernonia esculenta Hemsl., results show that: 30% ethanol group have hypoglycemic effect, but others not.In addition, and also 30% ethanol part of the fruit from Vernonia esculenta Hemsl. in mice adipose tissue insulin signaling pathways were studied, it was mainly investigated on the expression of five factors: PPARα mRNA, PPARγ mRNA, GluT4 mRNA and MnSOD mRNA, results show that: 30% ethanol extracts of the fruit from Vernonia esculenta Hemsl. can reduced the expression of PPARα mRNA of adipose tissue in db/db mice, and increased the expression of PPARγ mRNA, GluT4 mRNA and MnSOD mRNA.
