Mycobacterium tuberculosis can survive and proliferation in cells by escape immune surveillance and attack from macrophage. When the body’s natural immune system recognize the pathogen, the family of Toll like receptors play an important role in the body, after infection with Mycobacterium tuberculosis, TLR2 and TLR4 in macrophages and some cytokines have the function of immunoregulation Mycobacterium tuberculosis and antigen-presenting. At the same time, Mycobacterium can turn to regulate the expression of TLR2 and TLR4, if stimulate the TLRs continuatly, the body will produce a negative regulation, Mycobacterium tuberculosis will survival in the body long term. In recent years, NTM received widespread attention, and there are more and more research related to NTM, but the study on toll like receptors by macrophages to play anti NTM immune effect is less relatively.In this paper, At first we introduce the experimental data and its processing method. we detect the expression of TLRs and apoptosis of cells by flow cytometry in NTM infection at 0h, 4h, 12 h, 24 h, 48 h, 72 h, we detect the Changes in the levels of m RNA of TLR2 and TLR4 of RAW264.7 and Murine peritoneal macrophages by fluorescence quantitative PCR technology, and detect the expression of TNF-α using ELISA technology. Through making pathological section and acid fast staining sections, observed the lung and spleen pathological changes and bacteria distribution of rats after infected by NTM. The results showed: both M.gilvum and M.neoaurum infection can promote apoptosis of RAW264.7, and make the expression of TLR2, TLR4 of RAW264.7 and Murine peritoneal macrophages and TNF-α of RAW264.7 up-regulated, m RNA level of TLR2, TLR4 and TNF-α reached the highest expression in 4h, and the expression of TLR2, TLR4 and TNF-α were extremely increased compared with control group(P<0.05). And we found that the the up-regulated genes of TLR2 and TLR4 were positive correlation with the up-regulated expression of TNF-α, the lung and spleen of mice have pathological changes in a certain compared with the control group after infected with NTM. so we initial speculation that M.gilvum and M.neoaurum can promote apoptosis of RAW264.7, and make the expression of TLR2, TLR4 of RAW264.7 and Murine peritoneal macrophages and TNF-α of RAW264.7 up-regulated. the virulence of M.neoaurum is stronger than M.gilvum.At present, there are more and more research whic related to NTM, in this paper, several methods of the detection method mentioned above and the experimental results can provide the theoretical basis and reference for the experiment and research in the future. We can also through a deeper understanding of TLR2 and TLR4 to find the drugs anti NTM associated with TLRs, it may become a choice of the current treatment of NTM and have the important meaning to prevention and control the NTM.
