Objective:We analysed the expression of mice spleen TLR2, TLR4, CD4~+CD25~+Treg, and serum NF-κB to elucidate high-intensity exercise inducing mice spleen TLR2, TLR4 changing and its relation with NF-κB,CD4~+CD25~+Treg.Methods:Dividing randomly 100 Kunming mice into two groups, the control group(50) and exercise group(50);the control group contains 0week group, 1week group ,2week group, 3week group, and 4week group. The exercise group inclouds 0week group, 1week group ,2week group, 3week group, and 4week group. All the mice were fed in the same conditions, the control group received no interventions, but each exercise group swam with load about 5% their own weight according to the exercise design. We obtained mice spleen, thymus, and blood samples before exercise intervention and at the end of every week. Recorded the weight of spleen and thymus, used flow cytometry to detect the expression of mice spleen TLR2, TLR4, CD4~+CD25~+Treg, and analysed serum NF-κB by ELISA. The statistical software SPSS 17.0 was used for data analyse.Result:1.The spleen wight index increased at the first two week(sP<0.05), but declinded in the next weeks(P<0.05).It had no significance comparing to the controls; the thymus weight index decreased during four weeks, but also had no significance comparing to the controls.2.The expression of mice spleen TLR2,TLR4 increased from 2 week to 4 week in exercise group, and they reached top point in 2 week(P<0.01) . The expression of mice spleen TLR2,TLR4 was lower in 3 week and 4 week than in 2 week(P<0.05).There was no significantly changing in the control groups. 3.Serum NF-κB levels was higher in exercise group than in the controls during2week, 3week, and 4week(P<0.05,P<0.01). It reached top point in 3week and4week. There was no significantly changing in the control groups.4. The expression of CD4~+CD25~+Treg increased from 2 week to 4 week in exercise group, and they reached top point in 4 week(P<0.01) . There was no significantly changing in the control groups.Conclusions:1. 4 weeks load swimming delayed mice spleen growing, induced thymus atrophy quickly, and had negative effect on immune orgen developing and function.2. 4 weeks load swimming increased the expression of mice spleen TLR2,TLR4, elevated serum NF-κB level, and induced nature immune.3. 4 weeks load swimming induced mice spleen CD4~+CD25~+Treg increasing, Mediated adptive immune through TLR—CD4~+CD25~+Treg pathway.
