ObjectiveTo investigate the expression of orphan receptor GPR55 at transcriptional and translational levels in laryngeal squamous cell carcinoma tissues and adjacent tissues,and analyze the relationship between the expression level of GPR55 and different clinicopathological parameters.To observe the effects of small interfering RNA-targeting GPR55 on the metastasis and invasion of human laryngeal squamous cell carcinoma Hep-2 cell.MethodsThe expression of GPR55 was characterized respectively by reverse transcription-PCR 、 western blot and immunohistochemistry in 32 samples of laryngeal squamous cell carcinoma tissues and adjacent tissues,and the relationship between GPR55 expression level and different clinicopathological parameters was analysed.Three siRNAs-targeting GPR55 gene were synthesized.Laryngeal squamous cell carcinoma Hep-2 cell were instantaneously transfected using lipofectamine method.The mRNA levels of GPR55 in cells were determined with reverse transcription-PCR to choose the best si RNA-targeting GPR55 gene.The abilities of metastasis and invasion of transfected cells were assessed using transwell assay and wound-healing assay, respectively.ResultsThe expression of GPR55 was uniformly negative or weakly positive in adjacent tissues.Compared with the adjacent tissues, the expression of GPR55 was higher in laryngeal squamous cell carcinoma tissues,and the level of GPR55 was related with the differentiation of laryngeal carcinoma and rates of lymph node metastasis.No significant difference was found in the expression of GPR55 with different age,gender,clinical classification and PS scores.Transwell assay revealed that the number of Hep-2 cell transmembrane in siRNA-GPR55-3 group were significantly lower than normal control group and negative control group. Wound-healing assay revealed thatthe rate of Hep-2 cell migration in si RNA-GPR55-3 group were significantly lower than normal control group and negative control group in 12 hours and 24 hours.ConclusionThe expression of GPR55 was increased in laryngeal squamous cell carcinoma tissues,which have a certain relationship with differention of laryngeal carcinoma and lymph node metastasis.The siRNA-targeting GPR55 markedly decrease the abilities of metastasis and invasion in Hep-2 cell.
