| This article systematically studied on extraction, isolation, purification, separation and biological activities in vivo and in vitro of polysaccharides from roots of Cibotium barometz. The four crude polysaccharides were obtained from roots of Cibotium barometz by water extraction and base extraction. These polysaccharides were purified by DEAE-Cellulose 52, and Sephadex G-75, respectively. The gel permeation chromatography and could prove whether the polysaccharides were homogenous. The carbohydrate content of polysaccharide(CB50, CB70, CB90, CBB) were measured by phenol-sulfuric acid method. The homogenous polysaccharides were analysized by GC, HPLC and hydrolysis to study their monosaccharide composition. After carboxymethylation and sulfated modification of CB50, CB70, CB90, CBB polysaccharide, the yield and degree of substitution were calculated. The polysaccharide modified were compared with original polysaccharides in solubility and antioxidant activity of different aspects. Using ovariectomized rat model, we also studied the different parts of the polysaccharide against osteoporosis. |
