Objective: The study focused on preparing novel carboxymethyl chitosan(CMC)nanoparticles loading C-phycocyanin(C-PC) and exploring the effects of C-PC/CMC NPs on the growth of He La cells.Method: Three factors were chosen to design the orthogonal experiment: mass ratio between CMC and C-PC, the concentration of carboxymethyl chitosan(CMC) and the concentration of Ca Cl2. The optimal synthesized conditions of C-PC/CMCNPs were determined by the size and entrapment efficiency of NPs. NPs were prepared under the optimal conditions. C-PC, CMC, C-PC/CMCNPs were chosn to treat He La cells. CCK-8 assay was adopted to determine the effects on the growth of He La cells. The expression quantities of JNK, Bcl-2 and caspase-3 were determined by immunohistochemistry staining, immunofluorescence and Western blot, respectively.Results: The orthogonal analysis indicated that the optimal preparation conditions were as follows: weight ratio between C-PC and CMC was 1:2 and the concentration of CMC was 1 mg/ml and the concentration of Ca Cl2 was 1 mg/ml. The final entrapment efficiency C-PC/CMCNPs was 63.2% and the average particle diameter was 118.4 nm. All of C-PC, CMC and C-PC/CMCNPs could inhibit He La cells proliferation and induce He La cells apoptosis by up-regulating the expression of Caspase-3 gene and JNK and down-regulate Bcl-2. The antitumoral effect of C-PC/CMCNPs was the most significant.Conclusion: Satisfactory conditions to prepare C-PC/CMCNPs were obtained and optimal C-PC/CMCNPs were accepted with obvious sustained release character and anticancer effect. The anti-tumor effect of C-PC/CMCNPs might be achieved by inhibiting the growth of He La cells, promoting the transduction of apoptosis signals, and finally inducing cell apoptosis in the end. |
