Objective: To study the mechanism of vascular endothelial growth factor(VEGF)in bone marrow mesenchymal stem cells(BMSCs) induced by bone morphogenetic protein-2,It provides a theoretical basis for clinical prevention and treatment of osteoporosis and its complications for BMP-2.Methods: The osteoporotic animal model was established by performing ovariectom y on the 6-month-old female SD rats. 90 days after the bone mineral density of th e ovariectomized rats(the experimental group) and the rats(the control group) were measured. Under sterile conditions, the rat femur was isolated, and the osteoporosi s rats BMSCs were extracted and cultured with adherent method. Take the 4th gene ration cells osteogenic induction, and join the best concentration of rh BMP-2, and g roup intervention by adding with different concentrations of VEGF antisense oligode oxynucleotides(ASODNs), suramin and exogenous VEGF. observe the influence on alkaline phosphatase(ALP) activity After 2 days. statistics the number of mineralized nodule formation After 18 days.Result :1. The bone density of rats in control group was 0.179 + 0.006 g / cm2, while the bone density of rats in experimental group was 0.158 + 0.007 g / cm2. There were significant differences(P<0.05, t=4.180)..2. The formation of calcium nodules and ALP activity in the experimental group by the addition of rh BMP-2 were significantly higher than the control group.3. VEGF ASODNs group and Suramin group, the formation of calcium nodules and ALP activity were decreased gradually with the increase of dose. The concentration changes of exogenous VEGF group had little effect on the formation of calcium nodules and ALP activity.Conclusion :1. The bone mineral density of the 6-month-old female SD rats after the ovary treatment was decreased significantly.2. BMP-2 can promote bone marrow mesenchymal stem cells differentiating into osteoblasts.3. Exogenous VEGF alone could not promote osteogenic ability of osteoblasts, but it had no effect on BMP-2 promoting osteogenesis. VEGF may secrete some substances in autocrine form, and the substance influence the ability of BMP-2 promoting osteogenesis.
