Objective Impact factor (VEGF) protein-2expression in bone osteoporotic bone marrow stromal cells (BMSCs) into the observed in vitro osteogenesis and bone morphogenetic provide a new approach for the prevention and treatment of osteoporosis card..Method6to20months of age, body weight (300g±20g) SD bilaterally ovariectomized female rats, three months after the use of dual-energy X-ray absorptiometry to measure bone mineral density and body q compared with preoperative made to ensure that successful model, and the use of BMSCs morphology was observed under a phase contrast microscope bone marrow adherent cultured rat BMSCs osteoporosis, upside down.The rats were randomly BMSCs osteoporosis2nd generation (p2) cells were divided into experimental and control groups were added (with rhBMP-2) complete medium, liquid osteogenic bone induction.Two weeks later, Dasey Su Gong staining method to detect the formation of groups of cell calcium nodules, enzyme standard instrument measuring alkaline phosphatase activity and rt-pcr method to detect the expression of VEGF.Result (1) Whole body bone mineral density of rats:the whole body bone mineral density of rats before and after operation were0.179±0.007g/cm2,0.158±0.006g/cm2. Difference was statistically significant (t=4.180, P<0.05).(2)Alizarin red staining:BMSCs (P2) two weeks after the osteogenesis induced experimental dyeing effect is strong and the control group obviously.(3)Alkaline phosphatase activity: BMSCs (P2) of rabbits induced by two weeks after alkaline phosphatase activity of the experimental group is significantly higher than the control group, were15.62±1.27u/gprot,8.62±0.93u/gprot, difference was statistically significant (t=7.709, p <0.01).(4)BMSCs (P2) of rabbits induced the expression of VEGF in two weeks:the experimental group is significantly higher than the control group, were3.723±0.143,0.950±0.072. The difference was statistically significant (t=29.462, p<0.01).Conclusion RhBMP-2can improve to ovary osteoporosis rat BMSCs in vitro osteogenesis ability, the expression of VEGF can promote osteogenesis factor, regulate the expression of VEGF may be BMP-2to participate in one of the mechanisms of bone metabolism.
