Objective:The content of CD63, mast cell carboxypeptidase A3, terminal complement complex(SC5b-9)of the individuals were detected in the normal control group, anaphylaxis group to non-ionic contrast media and anaphylaxis group to beta-lactam drugs to analysis of these biological indicators, to investigate the mechanism of anaphylaxis to non-ionic contrast media and assess the significance of the basophil activation test by flow cytometry(BAT)in diagnostic.Methods:Dividing the samples into three groups, which contains 20 cases anaphylaxis to non-ionic contrast media, 40 cases anaphylaxis to beta-lactam drugs, 133 cases normal people(excluding various allergic diseases, coronary heart disease, craniocerebral injury,morphine overdose, etc). Collecting the peripheral venous blood with EDTA anticoagulation tube and ordinary vacuum tube, each tube keeps 2ml blood.(1) Which80μl whole blood is used for BAT, the flow cytometry and CD45/CD63/CD203 c antibody combination were employed to detected the CD63 expression of basophilic granulocyte in blood.(2) Collecting the serum to detected the levels of mast cell carboxypeptidase A3 with ELISA.(3) Collecting the plasma to detected the levels of SC5b-9 with ELISA.(4)All the data were analyzed and described by the SPSS 17.0 statistical software.Results:(1) The CD63 expression of basophilic granulocyte in the normal control group,anaphylaxis group to non-ionic contrast media and anaphylaxis group to beta-lactam drugs were 4.07±2.43 %, 4.07±2.43 % and 22.82±26.35 %, the results showed that the CD63 expression of basophilic granulocyte in group of anaphylaxis to non-ionic contrast media and group of anaphylaxis to beta-lactam drugs were significantly higher than that in normal control group(P<0.05), but there were no statistic significance between group of anaphylaxis to non-ionic contrast media and group of anaphylaxis to beta-lactam drugs.(2)The content of mast cell carboxypeptidase A3 in the normal control group, anaphylaxis group to non-ionic contrast media and anaphylaxis group to beta-lactam drugs were0.35±0.20 ng/ml, 0.35±0.20 ng/ml and 0.35±0.20 ng/ml, the results showed that the content of mast cell carboxypeptidase A3 in group of anaphylaxis to non-ionic contrast media and group of anaphylaxis to beta-lactam drugs were significantly higher than that in normal control group(P<0.05), but there were no statistic significance between group of anaphylaxis to non-ionic contrast media and group of anaphylaxis to beta-lactam drugs.(3)The content of SC5b-9 in the normal control group and anaphylaxis group to non-ionic contrast media were 0.13±0.05 ng/ml and 0.77±0.85 ng/ml, the results showed that the content of SC5b-9 in group of anaphylaxis to non-ionic contrast media were significantly higher than that in normal control group(P<0.05).Conclusion:(1) The increased levels of mast cell carboxypeptidase A3 showed that there are activation of mast cell in anaphylaxis to non-ionic contrast media.(2) The increased levels of SC5b-9 showed that there are activation of complement system in anaphylaxis to non-ionic contrast media.(3) There are activation of basophil both in group of anaphylaxis to non-ionic contrast media and group of anaphylaxis to beta-lactam drugs.(4) As a new kind of cell analysis technology, BAT also can be used to diagnose the anaphylaxis to non-ionic contrast media.
