Objective To oberve the changes of Digestion and absorption function and its effect on GK rats blood glucose and lipids after Duodenal-jejunal bypass, at the same time, To make a comparison with New Billiopancreatic diversion surgery( NBPD) group and sham surgery group to observe the blood glucose after lavaging glucose,sweet potato, egg whites,and to observe Fasting and 2-hour Postprandial blood lipid changes of the three groups rats and to explore the possible mechanisms involoved of operation treat type 2 diabetes.Methods 1. Thirty 12-week-old male GK rats According to the weight were randomized into three groups: respectively Undergoing Duodenal-jejunal bypass surgery(DJB group, n=10), New Billiopancreatic diversion surgery(NBPD group, n=10),sham operation(SHAM group,n=10). 2. Each group of GK rats, Body weight were detected 1week before surgery and1, 2, 4, 6 weeks after surgery in the fasting state and fasting plasma glucose(FPG), Random blood sugar were measured 2 week before surgery and2, 4, 6 weeks after surgery. 3. Oral glucose tolerance test(OGTT)were determined in 2 week before surgery and 2, 4, 6 weeks after surgery. 4. Fasting and 2-hour Postprandial Free fatty acid, Triglyceride, Cholesterol were determined before2 week surgery and 6 weeks after surgery. 5. 42 and 43 days after Surgery, using 50% glucose and sweet potato starch according to the 8 kcal/kg of quantity of heat to gavage, testing caudal vein blood at 0, 30, 60, 120, 180 min, and calculating the area under the curve(AUC) of blood glucose. 6. Postoperation 46 day, using egg whites lavage in accordance with the quantity of heatof 8 kcal/kg, then to test 0, 30, 60, 120, 180 min blood glucose levels,to calculate the area under the curve(AUC) of blood glucose.Results 1.pre-operation, three groups of rats no statistically significant difference were observed in body weight, 1 weeks after surgery, three groups of rats weight is lower than the pre-operation and there was statistically significant difference. 2, 4, 6weeks after surgery, there was no statistically significant difference between groups of rats. 2 week before surgery, and postoperative 2, 4, 6 groups of GK rats there were no statistically significant difference between fasting blood glucose. 2, 4, 6 weeks postoperatively, DJB group and NBPD group random blood glucose were lower than the sham group. 2, 6 weeks after surgery NBPD group and DJB group random blood glucose compared with sham operation group and preoperative, difference was statistically significant.6 weeks after surgery, NBPD group random blood glucose compared with sham operation group,difference was statistically significant. 2. preoperative OGTT, three groups rats of blood glucose area under the curve no statistical significance. 2, 4, 6 weeks after surgery, DJB group and NBPD group blood glucose area under the curve were significantly less than control group. 4, 6 weeks after surgery, NBPD group glucose area under the curv was statistically difference with sham surgery group. 3.2 week Before surgery, three groups rats fasting and postprandial 2h free fatty acids,cholesterol, triglycerides have no difference. 6 week after surgery, DJB group, NBPD group fasting cholesterol, free fatty acids compared with control, difference is statistically significant. DJB group postprandial 2h cholesterol, free fatty acids compared with control and pre-operation, difference is statistically significant. 4. Using sweet potato starch and glucose lavage, DJB group and NBPD group blood glucose area under the curve were lower than the sham group, both of the substances after lavage,NBPD group area under the curve compared with sham group, the differences were statistically significant. after Using starch lavage, DJB group and NBPD group 30, 60, 120 min blood glucose and blood glucose area under the curve are lower then Using glucose lavage.5. After egg white lavaging, three groups GK rats slightly fluctuations in blood glucose levels, no statistical difference was found between groups of blood glucose area under the curve.Conclusion 1. Both Duodenal-jejunal bypass(DJB) surgery and New Billiopancreatic diversion(NBPD)surgery, can improved GK rats blood glucose. 2. Both DJB surgery and NBPD surgery, can improved GK rats blood lipids. 3.GK rats After DJB surgery and NBPD surgery, The changes of digestion and absorption function are closely related with Billiopancreatic diversion, This may be one of the main mechanisms to improve blood glucose, blood lipids.
