Objective: To investigate the differential expression of Stat3 signaling pathway correlated cytokine IL-11 and Survivin in gastric cancer patients in order to understand possible roles that IL-11 and Survivin may play in carcinogenesis, progression and prognosis.Methods: Paraffin-embedded samples were obtained from 59 gastric cancer patients who underwent surgical operations at the First Affiliated Hospital Shihezi University School of Medicine, Shihezi, China, between January 1, 2004 and December 31, 2007. All of the patients were followed-up to April 1, 20014. Using tissue chip and immunohistochemistry Envision method to detect the expression of IL-11 and Survivin.Results:(1) The positive expression of IL-11 was 94.9% in GC, was higher than those in ANT(87.8%); and positive expression of Survivin in GC tissues was higher than those in ANT(96.6%/72.2%).(2) The expression of IL-11 was correlated with both depth of invasion and higher clinical staging; expressed Survivin was correlated with histological classification, depth of invasion, patients’ lymph node metastasis and higher clinical staging.(3) The high levels of IL-11 and Survivin were linked to poor prognosis in GC patients(P<0.001, respectively). The patients which both strong positive of IL-11 and Survivin have poor prognosis.(4) In GC tissues, a positive correlation was found in expressed levels between IL-11 and Stat3 activation, IL-6, IL-17 A, Survivin expression P<0.005, and negative correlation with SOCS3. Survivin was positive correlated with activited Stat3, IL-6, IL-11, IL-17 A expression P<0.005.(5) Cox multi-factorial regression analyses demonstrated clinical staging and the overexpression of IL-11 and Survivin being an independent factor predicting overall survival of GC patients.Conclusion: Overexpressed IL-11 and Survivin were correlated with poor prognosisof gastric cancer patients, and they may play a coordinated role in the development, progression and prognosis of gastric cancer.
