Research purposes:Detect the expression of PDGF, eIF4E in patients with uterine fibroids’fibroid tissue and its normal uterine smooth muscle tissue. Were compared the different expression in their menstrual cycle. Detect whether PDGF has a certain correlation with eIF4E when they express in uterine leiomyoma. Explore PDGF, eIF4E’s role in the development of uterine fibroids. In order to provide practical theory in early clinical diagnosis prevention and targeted therapy of uterine fibroids in the future. To some extent, it is good for the progress of the etiology study in uterine fibroids.Materials and methods:Randomly selected Yan’an University Affiliated Hospital’s 30 patients with uterine fibroids who is total abdominal hysterectomy in 2014.6~2014.12(Patients aged 41 to 50 years old). The fibroids are longitudinal cut and took about 1.0cm × 1.0cm × 0.2cm size of fibroid tissue in the central part of the tumor. It is confirmed is uterine leiomyoma postoperative pathology. ≧ 3 months without the use of hormone therapy before surgery. Take this study 30 cases of uterine leiomyoma, formalin solution soaked to the pathology department as the experimental group. Select the same specimen from normal myometrium fibroid tissue at least 2cm beyond 30 cases as the control group. In order to eliminate the effects of individual differences in hormone environment for experiments.First, the two specimens embedded in paraffin, making pathological HE staining. Slice clearly meets the test criteria, while identifying the different stages of endometrial. Re immunohistochemical SP method to observe the PDGF (AA), PDGF (BB) under a microscope, eIF4E in uterine leiomyoma and normal myometrium expression, as well as at different menstrual cycle, whether there are differences between the expression. Simultaneous analysis of PDGF (AA), PDGF (BB), eIF4E expression in uterine leiomyoma relevance. Brown particles appear in the cytoplasm of a positive reaction, capture images. Each slice of randomly selected five high magnification (x 400) vision, count the total number of positive cells and the total number of smooth muscle cells in the Vision, the percentage of positive cells obtained (positive cells / total number of smooth muscle cells in the calculation of total). The results of scoring (P):0 points:less than 5% of the fetal membranes trophoblast cells were positive; 1 point:5% - 10% of the fetal membranes trophoblast cells were positive; 2 points:11% - 50% of the fetal membranes nourish cells were positive; 3 points:more than 50% of the fetal membranes trophoblast cells were positive. And calculate the depth of the majority of positive cells based on staining intensity scores (I):3 points:strong coloration; 2 points:moderate stain; 1 point:a light coloration. The end result is integral histology (H)= P+I. The measured integral histology (H) organized into statistical data, using SPSS19.0 software for statistical analysis, all statistical indicators should be subject to normality and homogeneity of variance test. Of measurement data with normal distribution, using two independent samples t-test between the mean. The difference was statistically significant when P<0.05.When compare the correlation of expression of PDGF (AA),PDGF (BB) and eIF4E, using Pearson linear correlation analysis. The results:1、The expression of PDGF (AA), PDGF (BB), eIF4E in uterine leiomyoma and normal muscle tissuePDGF (AA), PDGF (BB) were positive expressed in uterine leiomyoma and normal muscle tissue. It’s brown particles mainly appear in the cytoplasm within fibroids. There is positive expressione of eIF4E in uterine leiomyoma, but there is almost no positive cells In normal muscle. Brown particles appear in the cytoplasm or nucleus of smooth muscle cells. The expression of PDGF (AA) in uterine fibroids is higher than the normal amount of smooth muscle (t= 2.36, P= 0.022); The expression of PDGF (BB) in uterine fibroids is higher than the normal amount of smooth muscle (t= 2.50, P= 0.015); The expression of eIF4E in uterine fibroids is higher than normal amount of smooth muscle (t= 3.26, P= 0.002). Differences between the three groups were statistically significant (P<0.05).2、PDGF (AA), PDGF (BB), eIF4E’s expression in the different menstrual cycle of uterine fibroidsThe expression level of PDGF (BB) in uterine fibroids secretory is higher than the proliferative phase (t= 2.52, P= 0.018), the difference was statistically significant; The expression of PDGF (AA),eIF4E is no significant difference in the secretory phase and proliferative phase(P>0.05).3、The correlation of PDGF (AA), PDGF (BB), eIF4E in uterine leiomyoma and normal smooth muscleThe Pearson linear correlation analysis showed that:The expression of PDGF (AA), eIF4E and PDGF (BB), eIF4E in uterine leiomyoma tissues is highly correlated, the correlation coefficients were r= 0.37 and r= 0.49, P<0.05. The expression of PDGF (AA),eIF4E and PDGF (BB), eIF4E is no correlation in normal smooth muscle, the correlation coefficients were r=-0.003 and r=-0.045, P>0.05. The expression of PDGF (AA) and PDGF (BB) are not correlated in uterine fibroids and normal smooth muscle, P >0.05, the difference was not statistically significant. The conclusioneIF4E is highly expressed in uterine leiomyoma, but did not express or little express in normal uterine smooth muscle tissue; The expression of eIF4E has not significant differences in patients with uterine fibroids proliferative and secretory phase; The expression of PDGF (AA), PDGF (BB) in uterine leiomyoma was higher than its adjacent normal muscle tissue;PDGF (BB) were significantly differentially expressed in patients with uterine fibroids proliferative and secretory phase in, and PDGF (BB)’expression levels in patients with uterine fibroids secretion was higher than the proliferative phase, and the expression of PDGF (AA) in uterine fibroids tissues proliferative and secretory phase no significant difference; The expression of PDGF(AA),eIF4Ein uterine leiomyoma tissues were significantly correlated, and showed a positive correlation, but no significant correlation in normal uterine smooth muscle; The expression of PDGF(BB),eIF4E in uterine leiomyoma tissues were significantly correlated, and showed a positive correlation, but no significant correlation in normal uterine smooth muscle; The expression of PDGF (AA),PDGF (BB) in the uterine fibroid tissue and normal muscle tissues were not significantly correlated.
