Renal Cell Carcinoma is one of the goals of human cancer tumor, its organizational manifestations appear lipid accμmulation. Studying the metabolic characteristics of Renal Cell Carcinoma can guide us explore its associated mechanisms.The main molecular function of malic enzyme(ME1 and ME2) is converting malic acid into pyruvate. Some recently report that ME1 and ME2 in cancer exist its functional role in tumor and its roles become more and more prominent, mainly in the liver, breast and throat cancer.However currently there is no features reports about ME1 and ME2 in kidney cancer.To explore the major role about ME1 and ME2 in renal cell carcinoma, we make the following experiment. Using the renal cell carcinoma tissues and the renal cancer cell lines, we want to known the different expression of ME1 and ME2 in renal cancer. The q RT-PCR assay method we used is to research the RNA level change of ME1 and ME2 in renal cancer tissues; the use of western blot assay to research the protein change of ME1 and ME2 in renal cancer tissues; the use of immunohistochemistry assay to detect the distribution and expression of ME1 and ME2 in kidney cancer slices; In addition, we also want to compare the changes of ME1 and ME2 between the normal kidney cell lines and the ki dney cancer cell lines. It was found that, compared with normal tissue,the expression of ME1 and ME2 in renal cancer tissues is reduced, but the reduced degree of ME1 is more apparent.In order to explore ME1 function in renal cell carcinoma, consistent wi th the organizational characteristics of renal tissues,the main used cell lines is the cancer cell line RC2, by viral packaging in 293 T and infection kidney cancer cell lines, prompting renal carcinoma cell line RC2 overexpressing ME1, then we detect cell proliferation,apoptosis and ROS level. It was found that, after overexpression of ME1,RC2 cell proliferation decreased significantly, and by flow cytometry test, ME1 overexpression in renal carcinoma cell line RC2 has no impact in apoptosis, while the cell cycle assay found that ME1 overexpression appears cell cycle arrest in G1 p Hase. And the ROS levels in the cancer cells were significantly different. The characteristics of kidney tumor tissue is consistent with the function of cell cycle arrest, ME1 can be further explored the mechanism of cell cycle regulation and the ROS product.The characteristics of kidney tumor tissue have something related to the function of cell biology of ME1.To futher research the mechanism of ME1 in renal cell carcinoma have specially significant.
