Objectives:Diabetic retinopathy(DR) are one of the most common and most serious diabetes complications, and it is one of the reasons of adults’ blindness.With the increase of number of diabetics around the world, DR is going to be the major cause of blindness and visual function damage.Try to find the relationship between the adiponectin(ADPN) and type 1 diabetes rat model of retinal tissue TGF- beta 1 and the ICAM- 1, and the relationship between each factor and the degree of diabetic retinopathy. Methods:Choose 40 inbreeding SD male rats(weight between 180g- 220g) at clean level, then divide them into 2 groups--the normal group(NC, n=16) and diabetes mellitus group(DM, n=24). Give diabetes mellitus group the intraperitoneal injection of STZ(65 mg/kg). Check the glucose of the rats 72 h after STZ injection,and glucose≥16.7mmol/L with the symptom of drinking more, polyuria, polyphagia symptoms, then the model is successful. Give normal control group equal amount of the intraperitoneal injection of potassium citrate buffer. Feed the two groups of rats for 1 week and observe. If the group one always meets the model of the standard, then don’t convert into the experiment. All give ordinary feed,observe the degree of retinopathy, the organization of TGF-β1 and ICAM-1 expression and the concentration of serum adiponectin of two groups of rats for 4 weeks to 8 weeks. Find the relationship between them according to the comparative analysis of the test conclusion. Results:1. The serum adiponectin level of Type 1 diabetes rats significant decreased compared to the level of the normal group. And with the progressing of the diseasing,the adiponectin of rat serum adiponectin level gradually declines.2. The expression of TGF-β1 and ICAM-1 between coursein organization gradually increased along with the development of course of disease.3. The degree of retinopathy increased gradually with the extension of type 1 diabetes duration. Conclusions:The serum adiponectin level of Type 1 diabetes rats is significantly lower than normal group. TGF-β1 expression in retina tissue increased significantly, and the fibrosis increased significantly. With the decreasing of adiponectin, the expression of TGF-β1 and ICAM-1 gradually increases, and the degree of retinopathy gets higher.
