Objectives:(1) To study the the pathogenesis of diabetic nephropathy by the proteomics approach.(2) To verify the expression levels of MBL associated serine protease 2 in different DN stages.(3) To discuss the relationship between mannose binding lectin with type 2 diabetic nephropathy.Subjects and methods:Part one: Four groups of age- and gender-matched subjects were enrolled in this study. Group A is the normal controls. Group B,C,D are type 2 diabetes patients whose urine albumin excretion rate(UAER) is less than 30mg/g creatinine, 30mg/g creatinine to 300mg/g creatinine, and more than 300mg/g creatinine respectively. Four groups of plasma protein were separated with two-dimensional electrophoresis.The data were analyzed with specified software in Image Master 2D Platinum(GE company). The proteins which expressed significantly differently among four groups were determined and then detected with Matrix assisted laser desorption/inionation time of fight mass spectrometry(MALDI-TOF MS).Part two: About 54 healthy unrelated subjects were recruited as control group(group A),and about 52 type 2 diabetes patients whose UAER is less than 30mg/g creatinine were recruited as group B, and about 30 type 2 diabetes patients whose UAER is 30mg/g creatinine to 300mg/g creatinine were recruited as group C,and 16 type 2 diabetes patients whose UAER is more than 300mg/g creatinine were recruited as group D.Serum levels of MASP2 in the four groups were measured with ELISE.The ELISE results would verify the results of the proteomics. Meanwhile, Serum levels of MBL were also measured with ELISE. Then to analysis thecorrelation between the MBL with MASP2 and between them with type 2 diabetic chronic complications.Results:Part one:The results of proteomics displayed that the expression of MASP2 elevated significantly in group B.Part two:(1) Compared to control group, serum levels of MASP2 in the type 2 diabetes patients were significantly higher(P<0.05).(2) Serum levels of MASP2 in group B and group C were higher than group C(P<0.05).(3) Serum levels of MBL in both of control group and experiment group had no statistically significance(P>0.05).Conclusions:(1) Serum levels of MASP2 elevated significantly in type 2 diabetic patients.(2) MASP2 may be closely correlated with the onset and progression of type 2 diabetic nephropathy.
