Basic Research On The Screening Of Effective Extracts From Vinegar Cooked Cyperus Rotundus L. For The Efficacy Of Resolving Liver-Qi

Cyperus rotundus L. was first recorded in Supplententary Records of Famous Physicians. It is the key medicine for gynecology, widely used for treating the stagnation of Liver-Qi in TCM clinic, and was named as the commander drug for healing Qi deseases and the marshal herb for gynecology. Been vinegar cooked, Cyperus rotundus L. have stronger effects of smoothing Liver to stop pain and resolving food stagnation than the raw one. Then it is good at treating hernia pain, bleeding by Qi-stagnation, and pain of gastral cavity. So vinegar cooked Cyperus rotundus L. was choosed as the model drug for the study of soothing Liver-stagnation.Based on the fuction of soothing Liver-stagnation, systematic solvent extraction was adopted to get 5 extractions as petroleum ether extraction, diethyl ether extraction, ethyl acetate extraction, n-butyl alcohol extraction and water extraction for gavage. Four animal models, acetic acid induced pain model, oxytocin induced dysmenorrhea model, blood stasis model of Liver-stagnation type and gastrointestinal dysfunction model of Liver-stagnation type, were applied to screen the effective extractions for soothing Liver-stagnation, by measuring related pharmacological indexes and stasis of indexes’ data with SPSS and Overall Desirability. Analysis of bivariate correlation was used to screen the indexes which has larger correlation with the fuction of soothing Liver, the count out the "Liver index" by Overall Desirability, to evalutate the effect of each extraction on soothing Liver-stagnation.The results showed that, petroleum ether extract, diethyl ether extract and ethyl acetate extract were effective extracts for soothing Liver to stop pain, petroleum ether extraction and ethyl acetate extraction were effective extracts for soothing Liver to regulate menstruation, stop pain, and disperse blood stasis. n-butyl alcohol extraction and water extraction were effective extracts for soothing Liver to regulate Qi to broad middle Jiao. The mouse body twisting times caused by acetic acid, mouse body twisting times and incubation period of oxytocin induced dysmenorrhea model, whole blood high shear reductive viscosity, plasma fibrinogen, and gastric remnant rate were the indexes having great relation with the fuction of soothing Liver. These indexes were used to work out the "Liver index" by Overall Desirability. petroleum ether extraction and ethyl acetate extraction were effective extracts for soothing Liver, which is agree with the results that essential oil is the effective part for Vinegar cooked Cyperus rotundus L.Through enhancing body’s tolerance and reducing body’s response to the pain, vinegar cooked Cyperus rotundus L. can soothing Liver to stop pain. Through lowering plasma fibrinogen in the blood and reducing blood viscosity, it can remove blood to disperse blood stagnation. By promoting gastrointestinal peristalsis, increasing gastric emptying and small intestinal propulsion, it can regulate Qi in the middle Jiao to help food digestion. The thesis successfully copied and applied Liver depression syndrome model of TCM to screen effective extracts of Cyperus rotundus L.. Through the statistical analysis method, imortant indexes were worked out, and "liver index" were calculated to evaluate the Liver-soothing effect of the extracts.
