Objective:Danggui Buxue Decoction had obvious curative effect on myocardial infraction, related with promoting the angiogenesis, but the mechanism remained unclear. This research studied the influence of Danggui Buxue Decoction on the expression of the vascular endothelial growth factor, the membranous receptor and the soluble receptor, revealing the mechanism that Danggui Buxue Decoction promoted angiogenesis in ischemic myocardial of myocardial infarction rats.Methods:Establishment of myocardial infarction model, after 24 hours,rats were randomized to sham operated group, model group, positive group, DBD high dose group, DBD low dose group. The next day, sham operation group and model group were given saline, three treatment groups received intragastric administration. After 4 weeks, rats were monitored the hemodynamic and were executed. HE staining observed the changes in myocardial tissue.Immunohistochemistry observed capillary density. VEGFmRNA,VEGFR1mRNA and VEGFR2mRNA were measured by Q-PCR, and their proteins were measured by Western blotting.Results:1.Hemodynamic test results showed:Compared with sham operated group, SW, CO, SV, Pes, EF, dP/dt max,-dP/dt min, dV/dt max,-dV/dt min significantly reduced (P<0.01), Ves, Ved, Ped significantly elevated(P<0.01), Tau significantly elevated(P<0.05) in model group. Compared with model group, SW, CO, SV, EF,-dV/dt min, dV/dt max significantly elevated (P<0.01), Pes,-dP/dt min significantly elevated(P<0.05),Ves significantly reduced(P<0.01),Ved significantly reduced (P<0.05) in three treatment groups.2.Immunohistochemistry showed that:The number of positively stained microvascular in three treatment groups were significantly higher than model group and sham operated group (P<0.01), model group was higher than model group(P<0.05).3.Q-PCR results showed that:The expression level of VEGF mRNA in three treatment groups was significantly higher than sham operated group (P<0.01)and model group(P<0.05); the expression of VEGFR1 mRNA in DBD high dose group and VEGFR2 mRNA in DBD low dose group was all higher than sham operated group and model group, the difference was satistically significant.4.Western blotting showed that:Compared with sham operated group and model group, VEGF protein expression was significantly higher in the three treatment groups (P<0.05); the protein expression of VEGFR1 in DBD high dose group and VEGFR2 in DBD low dose group were significantly higher (P<0.05). The protein expression sVEGFRl and sVEGFR2 in DBD high dose group group and DBD low dose group were significantly lower than sham operated group, model group and positive group (P<0.01).Conclusion:1.Danggui Buxue Decoction could improve the left ventricular function of the myocardial infarction rats.2.Danggui Buxue Decoction could enhance angiogenesis in ischemic area of myocardial infarction rats, the mechanism was that Danggui Buxue Decoction inhibited the expression of soluble receptor proteins, made a lot of vascular growth factors can combine with membranous receptor.
