Objective:To observe the clinical curative effect of acupuncture and placebo acupuncture for treating Insomnia with Anxiety-Depression state, and compare the difference between the two methods of acupuncture effect.Methods:The 95 subjects with Insomnia with Anxiety-Depression state were randomly divided into acupuncture or placebo acupuncture group. There are 48 cases who received ordinary acupuncture therapy in acupuncture group, and 47 cases who received placebo acupuncture in the other group. Steady state visual evoked potentials (SSVEP), Pittsburgh sleep quality index (PSQI), self rating Depression Scale (SDS) and self rating Anxiety Scale (SAS) were observed before and after acupuncture Operation in two groups of subjects, and then compared the difference between the two groups and within the group.Results:1.The total effective rate after the end of treatment:91.30% in acupuncture group and 9.09% in placebo acupuncture group.2. PSQI:(1) Inter group comparison: The total score of PSQI and each component scores were compared after the treatment and on the follow-up period in the two groups, with statistically significant difference (P < 0.05). (2) Inner group comparison:the total scores of PSQI and each component scores were lower than those before the therapy in acupuncture group, with statistical difference (P< 0.05). Comfort before and after acupuncture group treatment except sleep efficiency and compare the difference increasing dysfunction were statistically significant differences in the total score of PSQI and score, residual components of the project had no statistical significance (P> 0.05). In acupuncture group, the score of the sleep time, sleep efficiency, sleep disorders and daytime dysfunction on follow-up and after treatment,showed no statistical difference (P> 0.05); while comparison in PSQI total score, sleep quality and sleep time had statistical difference (P<0.05). Placebo acupuncture group on follow-up period and compared after treatment, statistically significant differences in sleep efficiency and sleep disorder (P<0.05), the difference score more than project showed no statistical significance (P>0.05).3. SAS and SDS: (1) Inter group comparison:difference of both SDS and SAS standard scores between two groups were statistically significant (P<0.05). (2) Inner group comparison:Both after treatment and in follow-up period, diffrences of SAS and SDS standard scores in acupuncture group were statistically significant (P<0.05), but in Placebo acupuncture group, there were no statistical significance (P>0.05). The difference of SAS and SDS standard score in follow-up period and after treatment had statistical significance (P< 0.05).4. SSVEP:(1) Inter group comparison:comparing all the SSVEP value between acupuncture group and placebo acupuncture group after treatment, the differences in Oz and O2 sites of Occipital lobe showed no statistical significance (P>0.05),and the other sites of the brain showed no statistical significance (P<0.05).(2) Inner group comparison: the potential value of each lobe leads differences were statistically significant (P< 0.05) in acupuncture group before and after treatment. Comparing the SSVEP value in Po5,Po6 sites of parietal lobe and Oz,02 sites of occipital lobe between the pre-treatment and post-treatment outcomes, it showed statistical difference (P< 0.05), but SSVEP value of the other sites of the brain showed no significant difference (P> 0.05).Conclusion:1. Acupuncture therapy can relieve both Insomnia and anxiety-depression symptoms of patients who suffered from insomnia with Anxiety-Depression state, and it also has certain long-term effect. While placebo acupuncture therapy showed neither effect of them.2. Acupuncture therapy can reduce SSVEP value of all the sites of the brain, Inhibitite the excitability of the whole brain.While placebo acupuncture therapy can only reduce SSVEP value of few sites.3. Acupuncture therapy is a safe and effective therapeutic method for treatment of insomnia with the state of anxiety and depression, and it is worthy of further promotion and carrying forward.
