Objective:To observe the antibacterial effects and heat clearing and blood activating activity of main compounds groups from Huanglian jiedu decoction (HLJDD), through in vitro antibacterial experiment and relieve fever experiment in vivo.Methods:1. By using the different concentration drug-containing ager plates, test the minimal inhibitory concentration (MIC) of major components groups from HLJDD and the component Phellodendrinechloride.2. To observe the rectal temperature, white blood cells, platelet and blood viscosity of septicemia rats after intravenous injection of Escherichia coli.3. Sprague-Dawley rats were randomly divided into control group, model group, aspirin group and HLJDD components groups. The rat model were established after the drugs were oral administration for 5 days. Besides the control group, the rats in another groups were firstly administrate with Carragenan i.p., and followed by LPS i.v. sixteen hours later. The rectal temperature, blood routine examination, marks of blood stream change and the levels of TNF-a, IL-6, ET-1, CGRP were observed to evaluation the antipyretic and blood activating activity. The levels of TNF-a, IL-6, ET-1, CGRP in serum are detected by enzyme linked immunoassay.Results:1. It shows a significant antibacterial activity of HLJDD and different components groups. Total alkaloids had the best inhibitory effects against gram-positive bacteria, the MICs was 8.37±5.05 mg/ml, and the inhibitory effects against gram-positive bacteria was significantly stronger than the inhibitory effects against gram-positive bacteria (MICs was 98.96±32.18 mg/ml,). The MICs of aqueous extraction against gram-positive bacteria and gram-negative bacteria were 43.53±27.53,88.54±39.60 mg/ml; while total flavonoids and total iridoids show poor effects on the most experimental bacteria, especially the total iridoids.2. All the compounds group could reduce the rectal temperature after administration drugs 2h. The water extracts of HLJDD could reduce the WBC, PLT level in the serum, meanwhile, total alkaloids and total iridoids could regulate the blood and viscosity.3. The rectal temperature, levels of WBC, whole blood viscosity, hematocrit, whole blood low shear rate, TNF-α、IL-6、ET-1 were all improved while the CGRP was decreased in rats. HLJDD compounds groups had a therapeutic action towards heat and blood stasis model, of which total flavonoids significantly down-regulated rectal temperature, total alkaloids ameliorated whole blood viscosity, hematocrit, whole blood low shear rate and also regulated the content of TNF-a and ET-1, remarkably.Conclusion:HJLDD showed strong antibacterial effects in vitro. Total flavonoids fraction, showing the best antibacterial effects, may the major material basis in HLJDD antibacterial effects. This investigation reveals that HLJDD has antipyretic activity, the total alkaloids and total flavonoids may be the possible material basis of this effect. The mechanism of this action probably relate to reducing the levels of TNF-a and ET-1.
