Objective:To study the correlation research of ESR1, FOXE1 gene single nucleotide polymorphisms(SNPS) and papillary thyroid carcinoma(PTC) and the curative effect of first iodine-131 ablation of thyroid remnants.Methods:(1).Using case-control study to research the clinical relevance of thyroid papillary carcinoma(PTC)and ESR1, FOXE1 gene single nucleotide polymorphisms(SNPS).(2). Selecting 93 PTC patients after total thyroidectomy to be ablationed by iodine-131 and to be followed up for 3~6months after ablation and the curative effect of short-term were evaluated. To analyze the influence of ESR1 & FOXE1 gene single nucleotide polymorphisms(SNPS), Age, sex, tumor TNM stage, tumor risk stratification, pre-treatment thyroglobulin(Tg) level and pre-treatment thyroid-stimulating hormone(TSH) level on curative effect of first iodine-131 ablation of thyroid remnants.Curative effect evaluation standard is divided into four level; cure, improve, did not change, worsen. One effective treatment = cure + improved+did not change,one uneffective treatment= worsen. Using SPSS13.0 software to Hardy-Weinberg equilibrium test, the χ2 test for R * C contingency table, the double variables of Pearson correlation coefficient test, the binary classification unconditioned logistic regression analysis.Results:(1)(1). There were significant statistics differences in the genotype and allele frequency of rs1867278A/C between the experimental group and the normal control group( χ2=9.029, P=0.011; χ2=9.123,P=0.003);(2). There were no statistics differences in the genotype and allele frequency of rs722207A/G、rs722209C/T、rs965513A/G、rs1867277A/G between the experimental group and the normal control group(P>0.05);(3).The genotype frequency of rs1867278A/C were 44.7%“C/G”and 55.3%“G/G” in normal control group. However, genotype frequency for rs1867278A/C was just only 100%“G/G”in experimental group.(4). The genotype and allele frequency of rs9340799A/G in experimental and normal control groups were out of accord with Hardy-Weinberg equilibrium(χ2=8.888,P=0.012<0.05;χ2=7.619,P=0.022<0.05)。(5). The genotype and allele frequency of rs11155816C/T、rs535364517A/G、rs538463552A/T、rs754078608A/G、rs535522715C/G、rs111482243C/T、rs531484350A/T、rs549601899C/G in experimental and normal control groups were all 100%“T/T”、“A/A”、“T/T”、“A/A”、“G/G”、“T/T”、“A/A”、“G/G”。(2)(1). Selecting 93 PTC patients after 3~6 months first iodine-131 ablation of thyroid remnants. 10 cases(10.7%)were cured,39(41.9%) improved,23cases(24.7%) were not change and 21(22.5%) ineffective.The total effective rate reached 77.4%(72/93). Single-factor analysis showed age,sex, tumor TNM stage, pre-treatment thyroid-stimulating hormone(TSH) level and rs965513、rs1867278 were associated with the curative effect of short-term on the first iodine-131 ablation of thyroid remnants(P<0.05). However,tumor risk stratification, pre-treatment thyroglobulin(Tg) level and rs9340799、rs722207、rs722209、rs1867277 were no significant effect on the curative effect of short-term on the first iodine-131 ablation of thyroid remnants(P<0.05). Binary classification unconditioned Logistic regression analysis indicated that tumor TNM stage, pre-treatment thyroid-stimulating hormone(TSH) level and rs1867278 were the main factors for the curative effect of short-term on the first iodine-131 ablation of thyroid remnants(P<0.05). Formula logistic regression equation was obtained as Y=6.187-2.87 x4(low-risk group)-2.546 x4( middle-risk group)-0.0423x6(pre-TSH level)-2.921 x13(rs1867278‘A/A’)-3.815 x13(rs1867278‘A/C’).Conclusions:(1)、(1).rs1867278A/C locus could be a clinicogenetic susceptibility loci in patients with PTC.(2). rs9340799A/G、 rs722207A/G、 rs722209C/T、rs965513A/G、 rs1867277A/G、rs11155816 C/T、rs535364517A/G、rs538463552A/T、rs754078608A/G、rs535522715C/G、rs111482243C/T、rs531484350A/T、rs549601899C/G locuses could not be clinicogenetic susceptibility loci in patients with PTC.(3).rs375606042A/G locus mutation genotype “C/G” could be a clinicogenetic susceptibility protective genotype in patients with PTC.(2)、(1). The curative effect of first iodine-131 ablation of thyroid remnants was so better after PTC surgery.(2).Age, sex,tumor risk stratification, pre-treatment thyroid-stimulating hormone(TSH) level and rs965513、rs1867278 were associated with the curative effect of short-term on the first iodine-131 ablation of thyroid remnants; tumor TNM stage, pre-treatment thyroglobulin(Tg) level and rs9340799、rs722207、rs722209、rs1867277 were no significant effect on the curative effect of short-term on the first iodine-131 ablation of thyroid remnants.(3). Synergetic effect of tumor risk stratification, pre-treatment TSH level and rs1867278 were the exact significant factors influencing the curative effect of short-term on the first iodine-131 ablation of thyroid remnants.(4). In this study, rs1867278 loci genotype "A/C" of FOXE1 gene could be the biggest influence factors influencing the curative effect of short-term on the first iodine-131 ablation of thyroid remnants.
