ObjectiveTo explore the effect of enriced environment on the expression of neurons and GSK3?,p-GSK3? protein in hippocampus region of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods7-day-old Sprague-Dawley rats were randomly divided into sham operation group,model group and enriched environment group,with 24 rats in each group.HIBD mouse model was prepared with reference to the latest modified Rice-Vannucci method.The rich environment group gave early massage therapy on the 7days.After the pups opened their eyes,they were given abundant environmental interventions.The sham-operation group and the model group did not have any special stimulation treatment and only given the same conditions.On the 14 days and 28 days after the intervention of rich environment,Morris water maze was used to evaluate the learning and memory abilities of rats in each group.Nissl staining was used to detect the survival of hippocampal neurons in each group.The expression of GSK3??p-GSK3? protein in area of the hippocampus was detected by Western Blot.Results1.on learning and memory ability:On the 14 days and 28 days after modeling,compared with the sham operation group,the time of escape latency of the model group increased significantly and the number of platform traversal decreased(P<0.05).Compared with the model group,the time of escape latency in the enriched environment group decreased significantly,and the number of platform traversal increased(P<0.05).2.Survival of neurons:Morphological and pathological changes of neurons in hippocampus of rats in model group were significantly changed on the 14th and 28th day after modeling compared with those in sham-operation group.Neuronal somatic bodies were sparsely arranged,irregularly shaped and neatly arranged,and nerve fibers arranged(P<0.05).Compared with the model group,the cell bodies of the hippocampus in the enriched environment showed a regular shape(circular,pyramidal,oval)arranged neatly and tightly,and the Nissl body Dense all showed normal,the difference was statistically significant(P<0.05).3.GSK3?and p-GSK3?protein expression:On the 14 days and 28 days after modeling,GSK3?protein expression and p-GSK3?protein expression in hippocampus in model group were significantly decreased compared with those in sham operation group(P<0.05).Compared with the model group,GSK3?protein expression was increased in the hippocampus of rats in the enriched environment group,while p-GSK3?protein expression was less in the hippocampus of rats in the enriched environment group(P<0.05).Conclusion1.Enriched environment can improves the ability of learning and memory in hypoxic-ischemic brain damage rats,and increase the survival of neurons in hippocampus,with neuroprotective effect.2.Enriched envirornment can upregulate GSK3?in hippocampus and down-regulate the expression of p-GSK3?,and effectively reduce hypoxic-ischemic brain injury.
