Human insulin-like growth factor 1 receptor(IGF1R) is a tumor growth promoting factor.It expresses highly in many malignant tumors. Long noncoding RNAs(lnc RNAs) is a general term for a class of RNA molecules over a lenth of 200 nucleotides. lnc RNAs are transcribed commonly in the genome. They regulate the expression of genes in a variety of levels in the form of RNA. A novel lnc RNA was found in the IGF1 R gene loci in a new research. It spans the promoter region of IGF1 R, and is in an antisense direction as compared to the IGF1 R coding m RNA.The lnc RNA was imprinted in normal human tussues, with monoallelic expression from the paternal allele and monoallelic silence from the maternal allele.It was named IRAIN(IGFI receptor antisense imprinted noncoding RNA). There has been no study on the expression levels of IRAIN in laryngeal squamous cell carcinoma in domestic and abroad so far. Objective to:To determine the expression level of a novel lnc RAN IRAIN in laryngeal squamous cell carcinoma and explore their correlation with the clinical and pathological features. The experiment is expected to provide new ideas and experimental basis to gene diagnosis and targeted therapy of laryngeal cancerMethods:25 patients with laryngeal cancer was selected who underwent partial or total laryngectomy at the Department of Otorhinolaryngology of the Second Hospital of Jilin University between February 2014 and February 2016. Total RNA was extracted from cancerous and adjacent normal specimens and c DNA was synthesized with RNA reverse transcriptase. The expression level of IRAIN was determined by q RT-PCR(real-time quantitative PCR) and their correlation with the clinical and pathological features was analyzed.The results were statistically analyzed by SPSS 19.0 software.Results:The expression level of IRAIN in cancerous specimens is significantly lower than that in adjacent normal specimens(P>0.05).The expression level of IRAIN had no significant relation with gender,age and TNM staging(P>0.05).The expression level of IRAIN had significant relation with tumor differentiation, whether there is lymph node metastasis and clinical classification(P<0.05).It decreased in patients with low degree of tumor differentiation,lymph node metastasis and supraglottic laryngeal cancer. Conclusions:1.The expression level of IRAIN in cancerous specimens is significantly lower than that in adjacent normal specimens,suggesting that IRAIN may be a antioncogene regulating the growth of laryngeal carcinoma cells.2.The expression level of IRAIN had significant relation with tumor differentiation, whether there is lymph node metastasis and clinical classification.It decreased in patients with low degree of tumor differentiation,lymph node metastasis and supraglottic laryngeal cancer.It suggests that IRAIN may act as a antioncogene to participate in the invasion and metastasis of laryngeal cancer.3.IRAIN was expected to be a biomarker with predictive value and the target of individualized therapy.
