The research background:For the perennial herb rhodiola rosea, alpine unpolluted zone growth in1800-2500 meters above sea level, the growth conditions, thus has strong vitality and special adaptability. Can fill gas clearing lung, educational yangxin,it is just a wide range of traditional Chinese medicine.Rhodiola glucoside acquired from alpine rhodiola precious ingredients,enhance immunity, eliminate depression, protection efforts, whitening, resist radiation, protect the nervous system damage repair etc.Man had two hippocampus, which located in the temporal lobe of left and right hemispheres. It is one part of the limbic system, has the function of improving memory and spatial orientation. The study found that in the AD(alzheimer’s disease, the human brain hippocampus is first of all, the area of damage, mainly characterized by memory loss and loss of direction of consciousness. There are some other diseases such as lack of oxygen to the brain anoxia and encephalitis can also lead to the hippocampus damage.Researchers believe that the hippocampus is closely related with learning,memory, organization structure, scientists in the study found that most of theneurons of the basal forebrain neurons in the brain to cholinergic neurons,raised from these neurons can directly onto the hippocampus, the cingulate cortex, and side to side, these structures are the main source of cholinergic innervation, one of the most important discovery is the content of the human brain acetylcholine are closely associated with memory formation and storage of Ach(Ach) is a Ch AT catalytic synthesis, and the ACh E decomposition, to bind to receptors with Ach and its biological effect, in the process of the whole neuromodulation in the human nervous system, there are two key enzymes involved in: Ch AT, ACh E, the two groups of key enzymes is through coordinating role to maintain the dynamic balance of Ach in the brain. In the two groups of enzymes, Ch AT is important indirect index to estimate the content of Ach in the brain, and its concentration is closely related to the content of Ach. In some scientific research, the scientists found that in the brains of elderly diabetes patients with cognitive impairment with Ch AT activity decreased, and the active and the degree of cognitive impairment of patients were positively related.Diabetic nerve system is one of the important complications of diabetes,light person in cognitive dysfunction, appear serious dementia, seriously affect the quality of life of elderly patients.Research purpose:Diabetes mellitus model of rat hippocampal CA1 area Ch AT and BDNF positive neurons of change and the influence of rhodiola glucoside.The research methods:Wistar rats were divided into three groups, each group of 10, were randomly divided into normal control group(CON group), diabetes(DM group)and rhodiola nucleoside group(DM + S group). Chain of DM and DM + S group were given urea with cephalosporins(STZ) intraperitoneal injection, the preparation of diabetes model. DM + S group was given rhodiola glucoside 50mg/kg lavage treatment. After 12 w, monitoring rats blood glucose and body quality change; Using Morris water maze observe its behavior change; Using immunohistochemical method to observe diabetes mellitus model of rat hippocampal CA1 area to Ch AT and BDNF positive neurons of change and the influence of rhodiola glucoside.The results of the study and significance:Rhodiola glucoside can increase diabetes mellitus model of rat hippocampal CA1 area to Ch AT and the expression of BDNF, promote the function of the central cholinergic system, adjust the survival, differentiation and regeneration of neurons, so as to improve diabetes rat model of learning and memory ability.
