Objective:To investigate the laryngeal squamous cell carcinoma p53 and MGMT gene promoter region of the 5’upstream sequence of single nucleotide polymorphisms and explore the p53 gene 5’upstream sequence consisting of two haplotype correlation with laryngeal cancer,we provid a genetic marker for the the incidence of laryngeal squamous cell carcinoma.Methods:We detected 96 cases of laryngeal squamous cell carcinoma and 102 control subjects of MGMT promoter region, four single nucleotide polymorphisms p53 gene 5’upstream sequence loci (rs1625649, rs2287499, rs2287498, rs228749) genotype using SNaPshot.We calculated and compared two groups of genotype and allele frequencies, applicated HaploView4.2 computing rs2287499, rs2287498, rs228749 values and haplotype frequencies and tested control subjects loci Hardy-Weinberg equilibrium.All the experimental data statisticed SPSS 17.0, chi-square test was used for statistical, p<0.05 for the difference was significant.Results:MGMT promoter region and p53 gene 5’upstream single nucleotide polymorphisms rs 1625649, rs2287499, rs2287498, rs228749 were polymorphic in the crowd.Four locus genotypes among patients and healthy controls no statistically significant difference (X2 values were 4.47,0.98,1.67,4.68, P> 0.05), MGMT promoter region locus rs1625649 allele frequencies between patients and healthy control groups were statistically significant (X2 value of 5.77, P<0.05) differences, p53 gene 5’upstream sequence locus rs2287499, rs2287498, rs228749 allele frequencies between patients and healthy control group the difference was not statistically significant (X2 values were 1.11,1.56,3.36, P> 0.05).Two haplotypes of rs2287499, rs2287498 and rs228749 between patients and healthy control group the difference was not statistically significant (X2 value 1.46, P> 0.05).Conclusion:MGMT gene polymorphism associated with the incidence of laryngeal cancer. |
