Radix Astragali, known as Huangqi in China, is commonly used in traditional Chinese medicine for treatment of many diseases, because it has many pharmaceutical effects such as improve immunity, detoxification, diuretic and sore myogenic. Mordern pharmacological studies have shown that Huangqi is immunostimulant, tonic, antiviral, anti-hypertensive, hepatoprotective and diuretic activities, etc. The clinically medicinal species are Mongolia Astragalus and Astragalus membranaceus, it is widely plated in China, mainly in Gansu, Shanxi and Inner Mongolia. Radix of the different orign has different quality, and different processing methods result that quality evaluation methods for Huangqi are insufficient to fully control the quality of the Astragalus. Therefore, the present atical investigated the different polarity fingerprint with High Performace Liguid chromatography through different polarity extraction solvent, and compared the Pharmacodynamics effects. Using statistical methods, study the spectrum-effect relationship based on pharmacodynamic experiments and determinate five ingredients of Radix with HPLC-DAD-ELSD to evaluate the quality from multi-angle, multi-faceted, and they can be experimental basis and technical supports for new quality control methods. The first part of this thesis is different polarity fingerprint and spectrum-effect relationship, and the second part is determination of five components of different Radix. The main research methods and results are as follows:The 10 batches Radix Astragali were collected from different areas in China and respectively extracted with 95% ethanol in refluxing extraction, which is the total extact The extract was dealed with liquid-liquid extraction in petroleum ether, ethyl acetate, ethanol and water in turn. Using High Performance Liquid Chromatographic method to establish the chromatographic fingerprints of the five extracts. Finding out the best part of effect in comparison with two pharmacodynamics experiment, diuretic and improving immunity. The ethyl acetate extract significantly improve the urine output of mice, in carbon clearance experiment, the phagocytic index of the mice with water extract is the largest, and the ability to enhance immunity is strongest The peak areas of all common peaks in HPLC spectrum were connected to the data of the effect by using grey relation statistic method. The spectrum-effect correlation was studied and the order of all component peaks’contribution for the effect was arranged.The 15 batches Radix Astragali were collected from different areas in China and respectively extracted with 95% ethanol in refluxing extraction, concentrated. Using High Performance Liquid Chromatography with HPLC-DAD-ELSD technology determinated the contents of Calycosin, Formononetin, Astragaloside, Astragaloside Ⅱ and AstragalosideⅢ. The results show that, Radix of the different orign has different contents of Calycosin, Formononetin, Astragaloside, Astragaloside Ⅱ and Astragalosidelll.The studies on different polarity fingerprint of Radix and spectrum-effect relationships and determination of multi-components will evaluate and contol comprehensively the quality of Radix, and provide a theoretical basis for the further development and utilization of the Radix.
