Aconite is the root of Aconitum carmichaeli Debx. It’s one of the common toxic traditional Chinese medicine. The mainly producing areas is Sichuan, Yunnan, Jiangyou is the genuine producing area. Aconite was contained in the "Shen Nong’s herbal classic", record as "the first product of reviving Yang for resuscitation". Fuzi has the effect of Yang deficiency, mainly for the death collapse, heart yang deficiency, cold dampnesspain and other symptoms. Modern pharmacological studies show that Fuzi has the effect of strong heart, anti-inflammatory and analgesic, anti-tumor. The clinical used of Fuzi in TCM was in treatment for all kinds of yang deficiency disease.Chemical components are complex in Fuzi, alkaloids are the main toxicity and effects components including diester-diterpenoid alkaloids, monoester-diterpenoid, aconine alkaloids and so on. There are many kinds of processing Fuzi and the quality is different from each other. So it is important to estabilish methods for simultaneous determination of alkaloids for the quality control of decoction pieces and patent medicine. And as it’s known to all that decoction and processing both have great impact on the components and clinical use.In view of the above problems, this paper mainly studies the following four parts:First part:The content of 13 alkaloids in Fuzi were determined. The results can show the difference of main toxic and effective constituents among commercial Fuzi more fully and accurately. It provided a scientific reference for processing, quality control and clinic application of Fuzi.Second part:This study employed UPLC/QQQ-MS-MS for the determination of 13 alkaloids in decoction of crude Fuzi and heishunpian, to study dynamic variation of the alkaloids during decocting. As a result for the crude Fuzi, content of diester-diterpenoid alkaloids decreased rapidly, and maintained low level after 4 h decocting. The monoester-diterpenoid alkaloids’ contents firstly increased and reached the peak in 4-6 h and then gradually deceased. Aconine alkaloids increased all the time during decoction process. Fuziline, songorine, karacoline and salsolinol firstly increased and reached to the peak at about 4 h and then tended to stability or reduced slightly. Trends of the 13 alkaloids were almost the same in crude Fuzi and Heishunpian, but the ranges of change in Heishunpian were observably narrower than crude Fuzi. After 2-4 h decocting, the contents of diester-diterpenoid alkaloids in crude Fuzi were at very low level which could ensure clinical safety and the total alkaloids were significantly higher than in Heishunpian decoction.Third part:The study explorded the dynamic changes of alkaloids in two kinds Fuzi by modern processing methods include steaming and baking. The results showed that the change trend of alkaloids by steaming was similar to decocting method. The content of Monoester-diterpenoid alkaloids and aconine by baking were significantly lower than that at the same processing time by steaming, and the decomposed rate of diester-diterpenoid was slower. These two kinds of processing methods have some differences, but they both can effectively remove the toxic components and retain the active ones, also they have the advantages of simple process, quality controllable. So these two processing methods were worth being popularized and applicated.Fourth part:A coupled method of macroporous resin and solid phase extraction to pretreatment was estabilished for the first time. And HPLC was employed to stimultaneous determine ester alkaloids in Chinese patent medicine. This methods can provide a reference for the quality control of patent medicine contains Fuzi.
