| Objective:Prepared by process optimization chitosan/glycerol phosphate,select the appropriate temperature-sensitive hydrogel concentration,temperature and pH value, in order to build a base for the graft CS,physical and chemical properties,biological toxicity,biocompatibility, as bone performance in line with the requirements of the clinical application of CS/GP thermosensitive hydrogel scaffold for antibiotic treatment of osteomyelitis provide new sustained release carrier material.Methods:By mixing chitosan,glycerophosphate,chitosan/glycerophosphate hydrogels, observed by an inverted test tube liquidity recording gel time,PH value and other physical and chemical indicators,and examines sodium glycerophosphate the amount of sodium affect the gel time of the case.On this basis, the preparation of the hydrogel contained antibiotics,the use of dynamic in vitro elution experiments to evaluate the in vitro release performance;in whole animal experiments, animal osteomyelitis model was prepared through the blood biochemistry examination,microbiological examina--tion,three-dimensional reconstruction of CT examination,pathological examination, further evaluation of the therapeutic effect of the gel contained syrup of osteomyelitis in rabbits.Results:The chitosan/glycerophosphate hydrogel when PH7.0 relatively stable, with the CS and NaHCO3 concentrations increased progressively shorter gel time, and with the increasing concentration of GP gel prolonged;Preparation of vancomycin chitosan/ glycerophosphate temperature-sensitive hydrogel delivery system, in vitro release experiments showed that:different doses of the drug contained in the gel, as vancomycin in vitro release rate of the drug the amount of increase slowed;60 days after surgery,HE staining mainly as there are a lot of model group sequestrum surrounded by inflammatory granulation tissue infiltration;the treatment group showed a large number of new bone formation, bone around the implant a little osteoblast, osteoclasts, osteoblasts osteoclasts simultaneously.Conclusion:Chitosan has a physiological PH/glycerophosphate hydrogel for in vivo studies used in vitro;vancomycin prepared hydrogels having certain in vitro dissolution rates of Staphylococcus aureus osteomyelitis model in rabbits have some treatment,indicating that this material has a strong potential for the treatment of osteomyelitis. |
