Objective: To observe the effect of Abnormal Savda Munziq(ASMq) on the endogenous metabolic changes in Plasma and Urine in depression rat model.explore the internal mechanisms of ASMq from the metabolic point of view.Methods: Depression model was established on 72 clean SD rats using chronic unpredictable mild stress(CUMs).three different dosages(1.5, 3.0, 6.0g·kg-1) of ASMq is given to the model group.Then, plasma samples and urine samples from all groups are analyzed by nuclear magnetic resonance(NMR)technology,and the dates of NMR spectra have been analyzed by Pattern recognition analysis to find out the changed endogenous metabolites in the rat`s plasma and urine samples Results: 1.Compared to the normal control group,Plasma levels of isoleucine,Lactic acid,creatine, glutamine, tyrosine, glycoprotein, lipid, unsaturated lipid, β- hydroxybutyric acid, acetic acid, alanine, α- and β-glucose are significantly increased(P<0.05),while plasma level of histidine is Significantly decreased(P<0.05)on the depression model group. compared to the control group,Plasma levels of isoleucine, lactic acid, creatine,glutamine, tyrosine, sugar, protein, fat, unsaturated lipids, β- hydroxybutyric acid,acetic acid, alanine, α- β- glucose and glucose are significantly increased(P<0.05) on the depression model group, while plasma level of histidine was significantly decreased(P<0.05). In contrast to the depression model group, plasma level of histidine, α-glucose, β-glucose are significantly increased(P<0.05) while plasma levels of carnitine, lactic acid, β-hydroxybutyrate butyric acid, creatine, pyruvic acid,phenylalanine, alanine, lipid(VLDL, LDL), unsaturated lipids, acetone, tyrosine,glycoproteins and acid content are decreased significantly(P<0.05) on all of the ASMq low, medium and high dose groups.in Plasma.2.Compared to the normalcontrol group,Plasma levels of Lactate, β-glucose, unsaturated lipids, tyrosine,phenylalanine are significantly increased(P<0.05),while plasma level of Isoleucine,β- hydroxybutyric acid, acetylsalicylic acid, pyruvic acid, α- glucose, creatine are Significantly decreased(P<0.05)on the depression model group.In contrast to the depression model group, urine level of Lipids(VLDL), isoleucine, valine, β-hydroxybutyrate, leucine, choline, glucoprotein, acetylcysteine, methionine,acetoacetate,pyruvic acid,citric acid,glutamine,α-,β- glucose,histidine,creatine,tyrosine, phenylalanine, are significantly increased(P<0.05) whil urine levels of lactic acid,carnitine, Trimethylamine oxide,glycine,unsaturated lipid,methanoic acid are decreased significantly(P<0.05) on all of the ASMq low, medium and high dose groups. Conclusion: abnormal savda munziq can adjust and improve the disordered glucose metabolism, lipid metabolism, amino acid metabolism of depression rat model at some extent. regulation of depression rats plasma metabolism and urine metabolites is the main aspects of the anti-depression mechanism of ASMq.
