Objective: To observe the effect of Banqiao dangshen Polysaccharide on cell proliferation, NO release, i NOs activity, IL-1β release, type II collagen expression in ATDC-5,Chondrocytes cell line —ATDC-5 cell were cultured in vitro.Methods: The method of water extraction and alcohol precipitation was used to extract Banqiao Dangshen Polysaccharide; CCK-8 assay was used to detect the proliferation of ATDC-5 cells cultured in vitro; ATDC-5 cells were divided into 5 groups, namely control group, LPS group, Banqiao Codonopsis pilosula polysaccharide group(low concentration group, middle concentration group, the high concentration group).Nitrate reductase method to detect the NO release; ELISA detection of IL-1 beta expression; Measure the expression of II type collagen expression with Western blot metheod.Results: 1 The extraction of Banqiao Dangshen Polysaccharide is 4.53g; 2 Some different concentrations of Banqiao Dangshen Polysaccharide act on ATDC-5 cell and the 1000 μg/ml of polysaccharide solution could inhibit the proliferation activity of ATDC-5 cells to a certain extent, compared with the blank group,( P<0.05), the difference is statistically significant; 3 Under the action of LPS, the release of NO and IL-1β,the activity of i NOs in ATDC-5 cells were increased, the difference is statistically significant compared to the blank group(P<0.05); But when adding Banqiao Dangshen Polysaccharide, the release of NO and IL-1β and the activity of i NOs in ATDC-5 cells were decreased significantly. 4 Under the action of LPS, the expressing of type II collagen in ATDC-5 cells was reduced significantly when compared with the control group, the difference was statistically significant(P<0.05), but Banqiao Dangshen Polysaccharide can resist the affect of LPS which inhibits synthesis of type II collagen in ATDC-5 cells.Conclusion: 1 LPS can stimulate ATDC-5 cells produce amounts of nitric oxide; 2 Banqiao Dangshen Polysaccharide can inhibited the release of NO in LPS induced ATDC-5 cell to some degrees(via inhibition of i NOs activity); 3 Banqiao Dangshen Polysaccharide can inhibit the expression of IL-1β in LPS induced ATDC-5 cell to some degree; 4 Banqiao Dangshen Polysaccharide can resist the affect of LPS which inhibits synthesis of type II collagen in ATDC-5 cells, which made the ATDC-5 cells can express relatively normal type II collagen when stimulated by inflammatory.
