Expression And Significance Of Adhesion Molecules CD62P,CD44 And CD54 In Peripheral Blood Of Infants With Bronchiolitis | | Posted on:2017-04-19 | Degree:Master | Type:Thesis | | Country:China | Candidate:W Wang | Full Text:PDF | | GTID:2284330485480399 | Subject:Pediatrics | | Abstract/Summary: | PDF Full Text Request | | ObjectiveDetection of adhesion molecules CD62 P, CD44 and CD54 in the peripheral blood of infants with bronchiolitis of acute phase and recovery phase,analysis the relationship between adhesion molecules CD62 P and CD44 and CD54,aims to investigate the expression and significant of adhesion molecules CD62 P and CD44 and CD54 in bronchiolistis.Materials and Methods 1 Research objectsSelected 33 cases of hospitalized children with bronchiolitis,include 33 cases which was in acute phase and 30 cases in recovery phase(33 cases of bronchiolitis in acute phase for 2 cases of early discharge, 1 case of specimens failed to difficult collection of qualified blood samples, so 3 cases lossed)who were hospitalized between September 2014 and May 2015.Selected 30 cases with bronchial pneumonia as bronchial pneumonia group and 26 cases of non infected children with indirect inguinal hernia, umbilical hernia, finger deformity as control group.Four groups are in line with the diagnostic criteria of "Zhu Futang practical pediatric science" [1],and the broncho pulmonary dysplasia, heart disease and immune diseases, parasitic infections were excluded. In this experiment, all the subjects did not take the glucocorticoid or anticoagulant drugs and immunosuppressants before 2 weeks ago. There was no statistical significance difference among each subject between age and gender. 2 Specimen collectionBronchiolitis in the acute phase group, bronchial pneumonia group and control group were fasting peripheral venous blood of 4 ml on admission day or the second day in the morning early,bronchiolitis recovery phase group on admission after5-7days,respectively.Get 2ml after citric acid 1:9 sodium anticoagulation to explore the expression expression of adhesion molecules CD62 P in peripheral blood. Get the upper serum of the other 2 m L venous blood without anticoagulant,which centrifugal separated at low speed of 15 minutes,frozen in-80 degrees Celsius refrigerator.The adhesion molecules CD44,CD54 levels in serum in 4 groups were detected after the specimen to be collected. 3 Experimental methodsThe adhesion molecules CD62 P positive expression percentage in the peripheral blood in 4 groups was detected by flow cytometry, and the adhesion molecules CD44, CD54 level in serum was detected using ELISA method.4 Statistical methodsUsing SPSS19.0 statistical software for statistical analysis of the data, The results were recorded by the mean plus ± minus standard deviation( x ±s).Test of normality and homogeneity of variance first,then using the single factor analysis of variance compare the groups, and the SNK-q test was used to compare the difference between the two groups. Correlation analysis using Pearson correlation analysis, ɑ equals to 0.05 is the test standard and P values less than 0.05 was statistically significant.Results 1 Results of adhesion molecule CD62 P in peripheral bloodThe level of the adhesion molecules CD62 P in the bronchiolitis group in the acute phase(35.5±9.9%)was significantly higher than those of bronchiolitis recovery group(12.7±2.2%;P<0.01), bronchial pneumonia group(27.0±10.7%;P<0.05)and control group(8.3±2.6%;P<0.01).The level of the adhesion molecules CD62 P in the bronchiolitis group in the recovery phase(12.7±2.2%)was also significantly higher than this in the control group(8.3±2.6%;P<0.01). 2 The results of the adhesion molecule CD44 in serumThe level of the adhesion molecules CD44 in the bronchiolitis group in the acute phase(147±25ng/L)was significantly higher than those in bronchiolitis recovery group(70±8ng/L;P<0.01),bronchial pneumonia group(112±22ng/L;P<0.05)and control group(46±6ng/L;P<0.01).The level of the adhesion molecules CD44 in the recovery phase(70±8ng/L) was also significantly higher than this in the control group(46±6ng/L;P<0.05). 3 The results of the adhesion molecule CD54 in serumThe level of the adhesion molecules CD54 in the bronchiolitis group in the acute phase(442±24ng/L)was significantly higher than that of bronchiolitis recovery group(238±12ng/L;P<0.01),bronchial pneumonia group(363±22ng/L;P<0.05)and control group(132±8ng/L;P<0.01).The level of the adhesion molecules CD54 in the bronchiolitis group in the recovery phase was(238±12ng/L),which was also significantly higher than those in the control group(132±8ng/L;P<0.01). 4 Correlation AnalysisPearson correlation analysis showed that the percentage of adhesion molecule CD62 P and CD44 levels were positively correlated in children with bronchiolitis in acute phase(r= 0.91, P< 0.05), and the serum of CD54 level is positively related(r= 0.95, P< 0.05), serum CD44 and CD54 were positively correlated(r = 0.72, P < 0.05), as shown in figure.
