This experiment was aimed at discussing the relevance between coronary heartdisease (CHD) and the expression of platelet collagen receptor glycoprotein Ⅵ (GPⅥ)by testing the expression of platelet GPⅥ and other platelet glycoproteins (CD62P,CD63), further studying the correlation between platelet activation and endothelial cellinflammation in CHD in vitro, providing certain theoretical basis for pathogenesis ofCHD.Objective: To investigate the expression of platelet GPⅥ, CD62P, CD63fromthe patients with CHD, and the correlation was analyzed among them; To investigatethe expression of intercellular adhesion molecule-1(ICAM-1) and vascuolar celladhesion molecule-1(VCAM-1) on the surface of endothelial cells after the interactionwith platelets from these subjects; To analyze the correlation between platelet GPⅥ andendothelial cell adhesion molecules; Finally to explore the clinical significance of aboveresearch.Methods: A total of97patients were admitted to the Department of Cardiology,the2nd Affiliated Hospital of Dalian Medical University from November2010toOctober2011. The subjects were divided into acute coronary syndrome (ACS) group(n=43), stable angina pectoris (SAP) group (n=21) and control group (n=33).5ml bloodsamples with citrate anticoagulation were collected prior for antiplatelet therapy. Theexpression of platelet GPⅥ, CD62P and CD63was tested by flow cytometry (FCM),then the correlation between platelet GPⅥ and platelet CD62P, CD63was analyzed.The inflammatory response markers of endothelial cells were also measured with theexpression of endothelial cell ICAM-1and VCAM-1by FCM, and the correlationbetween platelet GPⅥ and endothelial cell ICAM-1, VCAM-1was also analyzed. Results:1. Platelet GPⅥ, CD62P and CD63had more expression in the patients with ACSthan in SAP and control group (both P<0.05).2. In the CHD population, the expression level of platelet GPⅥ was significantpositively correlated with the degranulation markers of platelets (CD62P and CD63)(r=0.713and0.615, both P<0.05). The correlation analysis showed that the expressionof platelet GPⅥ was correlated with myocardial necrosis markers including troponin-I(Tn-I) and creatine kinase-MB (CK-MB)(r=0.325and0.272, both P<0.05) in the CHDsubjects, and also the expression level of platelet CD62P and CD63was correlated withplasma Tn-I (r=0.288and0.290, both P<0.05) and CK-MB (r=0.436and0.329, bothP<0.05).3. After the interaction of platelets from the different groups and endothelial cells,ICAM-1level of endothelial cells was found to be significantly higher in ACS groupthan in control group. While we failed to find the differences among different groupsregarding the expression of endothelial cell VCAM-1.4. In the CHD subjects, the expression of platelet GPⅥ was correlated with thelevel of ICAM-1in endothelial cells (r=0.371, P<0.05), while no correlation withendothelial cell VCAM-1was found.Conclusions:1. In ACS, the expression of platelet GPⅥ, CD62P and CD63was obviouslyincreased. Consequently, detecting the expression of platelet GPⅥ in the patients withCHD may be helpful for screening ACS. The expression level of platelet GPⅥ wassignificant positively correlated with platelet CD62P and CD63, therefore platelet GPⅥwas inferred to be one of the platelet activation indexes as well as CD62P and CD63.2. The expression of platelet GPⅥ, CD62P and CD63was correlated with plasmaTn-I and CK-MB. Platelet GPⅥ, CD62P and CD63might be inferred to be sensitiveindexes for indicating the severity of ACS.3. ICAM-1level of endothelial cells was found to be significantly higher after theinteraction with platelets in ACS, and it was correlated with platelet GPⅥ. Thereforethe conclusions were achieved that platelet GPⅥ in patients with coronary heart diseasemay be related to inflammatory reaction of endothelial cells.
