PART ONE:EXPRESSION AND CLINCAL SIGNIFICANCE OF SRX AND E-CADHERIN IN CERVICAL SQUAMOUS CELL CARCINOMA TISSUEObjective: To detect the expression of Sulfiredoxin and E-cadherin in normal cervical tissues(NC), high-grade squamous intraepithelial lesions(HSIL), and cervical squamous cell carcinoma(CSCC), and to investigate the role of Srx and E-cadherin in the occurrence, invasion and metastasis of cervical squamous cell carcinoma.Methods: Immunohistochemical SABC method was used to detect the expression levels of Srx and E-cadherin in specimens of 123 cases of cervical squamous cell carcinoma, 25 cases of HSILs, and 30 cases of normal cervical squamous cell epithelium. The correlation between Srx expression and E-cadherin expression and the correlation between the two proteins and clinicopathological characteristics of cervical squamous cell carcinoma were analyzed.Results: The expression of Srx was up regulated and E-cadherin was obviously decreased in CSCC tissues; the differences of the positive expression rate of Srx and E-cadherin in three groups were statistically significant, Srx and E-cadherin expression were negatively correlated in CSCC tissues. There was statistically significant difference in positive expression rate of Srx among cervical squamous cell carcinoma of lymph node metastasis and the infiltration of haemal tube, but Srx expression did not correlate with age, clinical stage(FIGO), volume of primarily tumor, the depth of cancer invasion and degree of differentiation. There was statistically significant difference in positive expression rate of E-cadherin among cervical squamous cell carcinoma of volume of primarily tumor, degree of differentiation and lymph node metastasis.Conclusions: The over expression of Srx in cervical squamous cell carcinoma is correlated with malignant clinicopathological characteristics, Srx expression is negatively correlated with E-cadherin expression, which indicates that Srx may promote development of cervical squamous cell carcinoma by regulating EMT.PART TWO:SRX MAY PROMOTE THE INVASION AND MIGRATION OF CERVICAL WQUAMOUS CELL CARCINOMA BY EMT AT THE CELLULAR LEVELObjective: To investigate the role of Sulfiredoxin(Srx) in regulate invasion ability and migration ability in Siha cells of cervical squamous cell carcinoma.Methods: Siha cells were selected in our study, and divided into three groups: The blank control group(Crontrol), negative control(Sh NT) and experience group(Sh Srx). The si RNA technology was used to reduce endogenous Srx expression in Siha cells of cervical squamous cell carcinoma. The interference effect of different treatment groups were detected by using Real-Time PCR and Immunocytochemistry. First of all, expression of E-cadherin and Vimentin were detected respectively by quantitative PCR method and Western blot method at the m RNA level and the protein level. Then Transwell invasion assay and cell scratch assay were used to detect the invasion and migration ability of Siha cells.Results: The results of Real-Time PCR and Immunocytochemistry showed that interference effect is remarkable in Sh Srx group of Siha cells comparing with the other two control groups(P<0.05). Meanwhile, the expression of E-cadherin was up-regulated in Sh Srx group both at at the m RNA level and the protein level(P<0.05). However, the expression of Mesenchymal marker, Vimentin, was down-regulated in Sh Srx group both at the m RNA level and the protein level(P<0.05). At last, we detect the invasion and migration ability, the results showed that compared with the two control groups, invasion and migration capabilties were suppressed sharply in Sh Srx group(P<0.05). However, we found that these results have no significant difference between the two control groups(P>0.05).Conclusions: Srx may promote the migration and invasion ability of Siha cells and may involoved in initiation and development of cervical squamous cell carcinoma by EMT.
