Objective:The purpose of this study was to investigate the effects of different intervention methods on adipogenic associated proteins of bone tissues in ovariectomized osteoporosis rats.Methods:Eighty female SD rats(3-month-old) were randomly divided into sham group and ovariectomized group.10 weeks after ovariectomy the ovariectomized group was divided into ovariectomized static group, estrogen group(E2,25μg/kg,3 times per week), lithium chloride group(LiCL,15mg/kg,3 times per week), genistein group(GEN, lmg/kg/day), treadmill group(18 m/minute,40min/day,5 uphill,4 times/week) and vibration group(90Hz, 0.5mm amplitude,15min every time, twice a day). After interventions for 8 weeks, detected the TC, TG level of serum and the change of bone biomechanical; detected femur bone microstructure by Micro-CT.In addition, the morphological changes of L2 were observed by HE staining and the protein expression of bone PPARy2, PPARβ, PGC-1 and FABP4 were detected by Western blotting.Result:After interventions for 8 weeks, 1)The PPARy2, PPARβ and FABP4 protein expression in bone tissue of genistein group were significantly higher than ovariectomized group, PGC-1 protein expression is lower than ovariectomized group; 2) The PPARβ, FABP4 protein expression of bone tissue in LiCl group were higher than ovariectomized group, the PPARy2, PGC-1 protein expression were significantly lower than ovariectomized group; 3) The PPARy2, PPARβ PGC-1 protein expression of bone tissue in treadmill group were significantly higher than ovariectomized group, the FABP4 protein expression was significantly lower than ovariectomized group; 4) The PPARy2, PGC-1 protein expression of bone tissue in vibration group were significantly higher than ovarieetemized group, the PPARβ, FABP4 protein expression were significantly lower than ovariectomized group.Conclusion:1)Small doses of genistein can partially improve the expression of PPARy2, PPARβ and FABP4 but inhibit the expression of PGC-1; 2) Lithium chloride can partially improve the expression of PPARβ and FABP4 but inhibit the expression of PPARy2 and PGC-1; 3) Moderate-intensity treadmill exercise can partially improve the expression of PPARy2, PPARβ and PGC-1 protein but inhibit FABP4; 4) The whole body vertical vibration can improve the expression of PPARy2 and PGC-1 but inhibit the expression of PPARβ and FABP4.
