Objective:As a chronic aging-related disease, postmenopausal osteoporosis has a seriously influence on the middle-aged and elder women. The research of prevention and treatment of postmenopausal osteoporosis has remarkably caused great attentions in our modern society.Using motion stimulation and drug treatment to prevent and to treat postmenopausal osteoporosis has became a supreme research. This research adopts the model of rats which be excised ovaries bilateral. To carry out the effects of treadmill exercise, body vertical vibration and related drug therapy for ovarian protein expression related to osteoporosis rats.Methods:Arranging eighty three-months-old healthy unfertilized female SD rats to divide randomly to SHAM and OVX. Making those rats recovery their vigor by restorative feeding for about 10 weeks after the operation, and divide randomly from OVX group to treadmill group, vibration, estrogen group, isoflavones gorse, lithium chloride group. To be specific, treadmill exercise is to continue for about 40 min,18 m/min speed, Angle of runway in 5,4 times per week.The group of vibration is decided to involve for about 15 min to 2 times a day,90hertz of frequency, vibration of amplitude float up and down in 0.5 cm,7 times a week. Using subcutaneous injection to estradiol group rats with dose of 25 mu g/kg of 17-beta estradiol, three times a week; Isoflavones gorse groups are measured in accordance with 1 mg/kg every day to fill the stomach,7 times a week. Using intraperitoneal injection to Lithium chloride group rats by of dose of 15 mg/kg, three times a week. Eight weeks later, analysis the changes of their weight every week,and then use HE staining to observe lumbar 2, on the right side of the proximal tibia paraffin histomorphology changes, with the TRAP staining to observe the change of the osteoclasts in bone marrow cavity, serum VEGF concentration was determined by Elisa, with Western blotting detection, lumbar spine, femoral, tibia bone marrow cells all the change of protein expression, using immunohistochemical detection of VEGF protein orientation changes.Results:(1) The weight of treadmill, whole-body vertical vibration, estradiol, genistein groups was significantly lower than that of ovariectomized group; (2) serum TRACP lithium chloride level in rats was significantly lower than that of ovariectomized group; (3) serum genistein group VEGF concentrations were significantly lower than that of ovariectomized group; (4)The treadmill, gorse, estradiol and lithium chloride can reduced significantly the ovary osteoporosis rat lumbar and tibia bone marrow cavity fat cavitation number;(5) The treadmill, the body vertical vibration, isoflavones gorse, estradiol, lithium chloride can reduced significantly to ovarian osteoporosis rats;(6) The treadmill, body vertical vibration, estradiol, isoflavones gorse, lithium chloride can promoted to ovary osteoporosis rat tibia, lumbar spine, bone marrow cells in expression of VEGF protein.(7) running platform, the body vertical vibration, estradiol, lithium chloride can promote to ovary osteoporosis rats femur the expression of VEGF protein, but the isoflavones groups had no that of effect.Verdicts:(1) The treadmill, body vertical vibration, estradiol, isoflavones can significantly restricted the gain of ovary osteoporosis rats weight;(2)Lithium chloride can significantly reduced ovary osteoporosis rats serum level of TRAP;(3) Isoflavones can significantly reduce to ovarian isoflavones gorse osteoporosis rats serum VEGF level;(4)The treadmill, isoflavones, estradiol, lithium chloride can significantly reduced ovary osteoporosis rat lumbar and tibia bone marrow cavity fat cavitation number;(5) The treadmill, body vertical vibration, isoflavones gorse, estradiol, lithium chloride can be significantly reduced to ovarian osteoporosis rats tibia bone marrow cavity, osteoclasts number;(6) The treadmill, body vertical vibration, estradiol, isoflavones, lithium chloride can promote to ovary osteoporosis rat tibia, lumbar spine, bone marrow cells the expression of VEGF protein.(7) running platform, the body vertical vibration, estradiol, lithium chloride can promote ovary osteoporosis rats femur the expression of VEGF protein, but had no effect on it.
