Objective:(1) To investigate the relationships between the AKT1 、 MDM2 gene polymorphismin and primary hepatic carcinoma.(2) Explore the interaction of AKT1 and MDM2 gene polymorphism in hepatic carcinoma patients and the environment on hepatic carcinoma. Methods:In case-control study, collect 101 cases primary hepatic carcinoma patients as case group, selected from primary hepatic carcinoma high-risk groups 81 cases as intra- control group, at the same time, collect 102 case of healthy people as extra-control group. Through the self-made questionnaire of hepatic carcinoma epidemiological investigation to collect the medical records. PCR-RFLP was used to detect the gene polymorphism of AKT1 and MDM2. Result:(1)there were no significant differences of gender, nationality, age distribution. Cultural degree, marital status,occupational exposure history, per capita income, ever suffer from second liver, fatty liver,presence of liver cancer and liver cirrhosis, diabetes and family history of primary hepatic carcinoma have exist differences in three groups, the difference was statistically significant.rs1130214 each genotype and allele distribution difference between three groups, rs3730358 each genotype and allele distribution have difference between the three groups, rs2279744 each genotype and allele distribution hava difference between the three groups, the difference was statistically significant(P<0.05). In order to gene mutation or not as dependent variable, Logistic regression showed that the person who is primary hepatic carcinoma and primary hepatic carcinoma high-risk groups are more likely to mutate in rs1130214 of AKT1 gene(OR=1.91,95%CI:1.25~2.91), the same with male(OR=2.70,95%CI:1.66~5.00)(OR=2.70,95%CI:1.66~5.00).(3) GMDR analysis shows that Gene-Gene interaction model has no statistical significance. GMDR analysis results show that Gene-environment interaction there are statistically significant interaction model is(hepatitis b, diabetes),(hepatitis b, diabetes,),(rs1130214, hepatitis b,diabetes, and sleep time), the above interaction can affect the incidence of liver cancer.Conclusion:rs1130214 AKT1 gene gene polymorphism have a certain relationship with the occurrence of primary hepatic carcinoma; rs1130214, rs2279744 exist interaction on the occurrence of cancer of the primary hepatic carcinoma;There is interaction betwee Rs1130214 gene polymorphism with environmental factors.
