| Objective: To analyze the expression of HSP27, SM22, TAGLN2 and P38 MAPK in esophageal squamous cell carcinoma of Kazak and Han nationalities, According to the clinical pathological parameters to screen molecular markers. Methods: To use Western Blot technology in 28 pairs of fresh esophageal squamous cell carcinoma tissue analyzed the expression of HSP27, SM22, TAGLN2 and P38MAPK; Immunohistochemical method was used to detect the expressions of HSP27, SM22, TAGLN2 and P38 MAPK in 162 cases of ESCC and 50 cases of normal esophageal tissues, and their correlations to the clinicopathologic characteristics of ESCC were analyzed; ROC curve to evaluate the diagnostic value of each protein; Cox regression analysis of prognostic factors associated with esophageal squamous cell carcinoma. Results:(1) Western Blot found that the expression of the HSP27, SM22, TAGLN2 and P38 MAPK in fresh esophageal squamous cell carcinoma tumor tissue than adjacent normal tissues(P<0.05); HSP27 and SM22 in esophageal squamous cell carcinoma with lymph node metastasis was higher than that in the group without lymph node metastasis(P<0.05);(2) Immunohistochemistry verified the expression of differences, suggesting that the positive expression rate of 4 protein in cancer tissues was higher than that of normal mucosa(P<0.001); Calculation of AUC by ROC, TAGLN2 have a higher diagnostic value(AUC=0.902, P<0.001), The clinical pathological parameters analysis: Expression of HSP27 positive and lymph node metastasis correlation(P<0.05); SM22 expression positive and infiltration depth(P<0.05), lymph node metastasis(P<0.05), there is a correlation; TAGLN2 expression positive and Kazak(P<0.05), infiltration depth(P<0.05), lymph node metastasis(P<0.05) there is a correlation; p38 MAPK expression positive and infiltration depth(P<0.05)there is a correlation;(3) Cox regression model for survival analysis showed that lymph node metastasis, infiltration depth, HSP27 Expression were correlated with survival time. Conclusion: HSP27, SM22, TAGLN2, P38 MAPK is an important molecular changes in the development of esophageal squamous cell carcinoma; TAGLN2 and SM22 can be used as molecular markers for diagnosis of esophageal squamous cell carcinoma; HSP27 can be used as markers for prognosis evaluation. |
