Objectives:In recent years,the relationship of between HSPs and cancer is the hot areas in cancer research, including HSP70 and HSP27 are closer with tumor. Studies found that the expression of HSPs increase in a variety of tumor cells, And it is related to the apoptosis of tumor cell and tumor immunity, etc. The expression level of HSP can also be detected in the serum of cancer patients,except in tumor cells and tissues. Extracellular HSP as a biological activated factor plays a series of biological functions,and plays an important role in the developed process in a variety of tumor diseases.Studies suggest that to some extent the intracellular HSP has a important role in the regulation of apoptosis of tumor through between mediated anti-apoptotic pathway activated and inhibits the pro-apoptotic pathway, promoting the expression of HSP,and inhibits the apoptosis of tumor cells.eHSP can regulate the body's tumor immunity, and break the immune tolerance of tumor,and induce the body's immune system to produce tumor cell-killing. It has shown in the vitro studies that, extracellular HSP70 can play a role in anti-tumor,through stimulating dendritic cells to synthesis and release the pro-inflammatory cytokines,activating natural killer cells, and enhancing the cellular immune function. Reported in the literature abroad immunohistochemical ELISA method was used to reseach the serum HSP70 antibody levels of ESCC, stomach cancer patients and normal healthy subjects. The study found that the serum HSP70 antibody levels of patients with ESCC were significantly higher than that of gastric cancer or colon cancer or normal healthy subjects. Studies suggest that HSP70 antibody not only as a marker of cancer used to diagnose, but also as a new and effective method of immunal-therapy to guide the treatment of cancer. Esophageal cancer is one of the most common malignant tumor in China,the regional differences of incidence rate is significant,Hebei province is the higher incidence f esophageal cancer in China, the average annual mortality rate is up to 303.37/1000000 in individual region, it ranks in the first place in the world The main pathological type is squamous cell carcinoma.But the serum concentration of Hsp27/Hsp70 of patients with esophageal squamous cell carcinoma and the relation between the serum concentration of Hsp27/Hsp70 and the clinical pathological features, the infiltration of NK cells and DC, there are few international researches in this regard. We plan to use the immuno-histochemical ELISA method in the topic to assay the expression levels of preoperative serum HSP27 and HSP70 of patients with ESCC of quantitative analysis of the situation, and Studied the relationship between the clinicopathological features According to the expression level,we divide it into both high and low groups,study the relationship between the serum expression of HSP27?70 and the clinicopathological features. Two-step immunohistochemical technique was used to detect the infiltration of NK cells and DC in ESCC,Reseach the relation of between the expression concentration of heat shock protein and the infiltration of NK cell and DC, to look forward to understanding the possible effection about HSP27 and HSP70 in the process of oesophagus canceration to the immunal system. To understand the relationship between the HSP27/70 in the developed process of ESCC and non-specific tumor immunity.Methods:1 31 cases of ESCC patients were selected from May 2009 to August 2009, Thoracic-surgical-department, Fourth Hospital of Hebei Medical University. 20 cases were male, 11 cases were female, aged between 43 ~ 71 years old, the average age is 61-years-old, all patients were not given the radiotherapy and chemotherapy, without complications and no fever. 20 cases healthy persons in good health examed by our hospital examination center were selected, 11 cases were male, 9 cases were female, aged between 27 ~ 67 years old, the average age is 58-years-old. All of the subjects with fasting were distilled 3ml venous, extract the serum after be centrifugaled, to detect the serum concentration of HSP27 and HSP70.2 Using the ELISA to detect HSP concentrations of serum in the experiment, and read the OD value of each hole at a wavelength of 450nm by microplate reader and the concentration of HSP by the computer system software.Using the two-step immunohistochemical technique to test the infiltration of NK and DC in esophageal squamous cell carcinoma, Negative control with PBS instead of the first antibody.Results:1. The comparison of the ESCC group and the healthy control group in between gender and age.The Chi-square test results showed there is no statistical significance in the two groups in term of gender (P> 0.05). The Rank-sum test results showed there is no statistical significance in the two groups in term of age (P> 0.05).2.. The contrast in the aspect of serum concentration of HSP27/70.between the two groupsThe statistical analysis results showed the maximum serum concentration of HSP27 in ESCC patients is 9.96988,the minimum serum concentration is 2.09891,the median serum concentration is 3.69993. the maximum serum concentration of HSP27 in healthy control group is 3.83198,the minimum serum concentration is 1.20591,the median serum concentration is 2.22345. the maximum serum concentration of HSP70 in ESCC patients is 6.31761,the minimum serum concentration is 1.26516,the median serum concentration is 1.87215. the maximum serum concentration of HSP70 in healthy control group is 1.94051,the minimum serum concentration is 1.00988,the median serum concentration is 1.44268. The serum concentrations of HSP27/70 in patients with ESCC was higher than of healthy control group.There is statistical significance in the serum concentrations of HSP27/70 between the two groups (P=0.000).3 The relationship between the serum concentration of HSP27/70 in patients with ESCC and clinical pathological features.The concentration of HSP were divided into high and low concentrations according to the distribution trend of concentration.14 cases (5 cases were male,9 cases were female)of high concentration of HSP27 and 17(9 cases were male,8 cases were female) cases of low-concentration group in the ESCC group,16(11 cases were male,5 cases were female) cases of high concentration of HSP70 and 15(9 cases were male,6 cases were female) cases of low concentration group in the ESCC group.3.1 The relationship between the serum concentration of HSP27 in patients with ESCC and clinical pathological features.The Rank-sum test results showed there is not statistical significance between the high and low serum concentrations groups of HSP 27 of patients with ESCC and different tumor location, length of tumor,depth of tumor invasion, pathological stage of tumor and metastasis of lymph node (P>0.05).3.2 The relationship between the serum concentration of HSP70 in patients with ESCC and clinical pathological features.(1) The Rank-sum test results showed there is no statistical significance between the high and low serum concentrations of HSP 70 of patients with ESCC and different tumor location,depth of tumor invasion and metastasis of lymph node (P>0.05).(2) The Rank-sum test results showed there is statistical significance between the high and low serum concentrations of HSP 70 of patients with ESCC and different length of tumor and pathological stage of tumor (P﹤0.05).①The relationship between the serum concentration of HSP70 in patients with ESCC and different length of tumor.The high and low serum concentrations of HSP70 of patients with ESCC is no statistical significance between the tumor length of≤3cm group and 3 ~ 5cm group(P>0.05). The high and low serum concentrations of HSP70 of patients with ESCC is statistical significance between the tumor length of≤3cm group and≥5cm group(P﹤0.05). The high and low serum concentrations of HSP 70 of patients with ESCC is statistical significance between the tumor length of 3~5cm and≥5cm group(P﹤0.05).②The relationship between the serum concentration of HSP70 in patients with ESCC and different pathological stage of tumor.The high and low serum concentrations of HSP70 of patients with ESCC is not statistical significance between the phaseⅠgroup and phaseⅡgroup(P>0.05). The high and low serum concentrations of HSP 70 of patients with ESCC is statistical significance between the phaseⅠgroup and phaseⅢgroup(P﹤0.05),the phaseⅡgroup and phaseⅢgroup(P﹤0.05) also.4 The relation between the serum concentrations of HSP 27/70 of patients with ESCC and the infiltration of NK cells and DC in the corresponding tissue.The Rank correlation analysis showed there is no correlation in the analysis between the serum concentrations of heat shock protein 70 of patients with esophageal squamous cell carcinoma and the infiltration of NK cells and DC in the corresponding tissue, There is no statistical significance.Conclusion: 1 There is no significant difference in the aspect of gender and age between the ESCC group and the healthy control group.It tips that the serum concentration of HSP27/70 in the two groups does not vary according to the change of gender and age.2 There is obvious differences in the serum concentrations of HSP27/70 between patients with ESCC group and the healthy control group,It tips that the volume of HSP27, 70 released into the blood gradually increase with the progress of ESCC.3 The concentration of HSP were divided into high and low concentrations according to the distribution trend of concentration.There is no significant difference between the high and low serum concentrations groups of HSP27 of patients with ESCC and different tumor location, length of tumor,depth of tumor invasion, pathological stage of tumor and metastasis of lymph node, It tips that the clinical and pathological features of ESCC has no effect on the volume of the HSP27 release into the blood.There is no significant difference between the high and low serum concentrations of HSP70 of patients with ESCC and different tumor location,depth of tumor invasion and metastasis of lymph node.There is no significant difference between the high and low serum concentrations of HSP70 of patients with ESCC a and different length of tumor and pathological stage of tumor. It tips that the length and pathological stage of tumor affect the volume of HSP70 release into the blood.The volume of HSP70 released into the blood gradually increase with the progress of ESCC,especially the length and pathological of tumor.4 There is no correlation in the analysis between the serum concentrations of HSP70 of patients with esophageal squamous cell carcinoma and the infiltration of NK cells and DC in the corresponding tissue, it tips that the relationship between the serum concentration of HSP27/70 in ESCC and the innate immune cells in which NK cells and DC act the main effect to be studied further.
