Ganoderma luciderm is a well-known medicinal fungus which belongs to the family of Polyporaceae. In China, it has been used as a traditional Chinese herb for more than two thousand years. JUNCAO Ganoderma lucidum which was cultivated by JUNCAO technology has medicinal properties of other Ganoderma luciderm. What’s more, compared with ordinary basswood Ganoderma luciderm and wild Ganoderma luciderm, its effective composition content is higher. It has been shown that Ganoderma lucidum polysaccharides, as part of the most important functional components, have great effect on tumor inhibition, immunomodulatory, scavenging oxygen free radicals, lowering blood sugar, lowering blood fat and protecting the liver, etc. JUNCAO Ganoderma lucidum was regarded as raw materials in this experiment. Ganoderma lucidum polysaccharides were extracted from the fruiting bodies of Ganoderma lucidum, and the water-soluble polysaccharides were isolated and purified. Their chemical structure and antioxidant activity of Ganoderma lucidum polysaccharides and TP compounds were investigated by polymer physics and biological test methods.1. Water-soluble polysaccharides from JUNCAO Ganoderma lucidum fruiting bodies were extracted by hot water, and the total sugar content of Ganoderma lucidum polysaccharides was determined. The optimum of water-soluble polysaccharides extraction was established by single factor experiment and orthogonal experiment. The results indicated the optimal extraction conditions were extraction time of 3 h, temperature of 100℃, the solid to liquid ratio of 1:25, respectively. In this way, the highest yield of polysaccharides was 2.38%.2. When using the Sevag method and PA method to remove protein, the results showed that the PA method was more suitable for removal of the protein in Ganoderma lucidum polysaccharides by comparing protein removal rate and polysaccharide loss rate. After removing pigment by AB-8 resin and dialysis off small molecules, water-soluble Ganoderma lucidum polysaccharides extracted by hot water was isolated from JUNCAO Ganoderma lucidum fruiting bodies by DEAE Sephadex A-25 chromatography, and four components, respectively named as GLPS-1 the GLPS-2, GLPS-3 and GLPS-4, were obtained.3. The homogeneity of GLPS-1, GLPs-2, GLPS-3, and GLPS-4 were evaluated by Sephadex G-200 chromatography. The results showed that GLPS-1, GLPS-2 and GLPS-3 were all homogeneous components, but GLPS-4 might be more complex polysaccharide fraction.4. By the analysis of infrared spectroscopy and 1H NMR spectra, it was found that three JUNCAO Ganoderma lucidum polysaccharides components, GLPS-1, GLPS-2, and GLPS-3, showed the characteristic absorption peaks of typical carbohydrates. The sugar residues of GLPS-1, GLPS-2 and GLPS-3 were pyran ring, the connecting way of glycosidic bond might be the β-type.5. All the components of JUNCAO Ganoderma lucidum polysaccharides had good removal effect on hydroxyl radicals and DPPH free radicals, and their reducing power was also strong. In terms of total antioxidant capacity, the effect of the crude polysaccharides was significantly stronger than the purified polysaccharides. While in the composite antioxidant effect of JUNCAO Ganoderma lucidum crude polysaccharides with TP, antioxidant capacity of compound (removal effect on hydroxyl radicals and DPPH free radicals, and total reducing power) showed significant synergy. Among them, the synergistic effect of the compound removing hydroxyl radicals was the most significant, and it could be increased to a maximum of 15 times.
