Objective: To observe changes of APTT,PT,TT and the blood coagulation factors activity(FⅡ、FⅤ、FⅧ、FⅨ、FⅩ、FⅪ、FⅫ)in rabbits with different doses of dabigatran etexilate to help us understand the effect of dabidatran on coagulation system and its mechanism, also hope to have certain guiding significance for clinical medication.Materials and methods: select 36 healthy experimental rabbits with no significant differences in age, size, body weight and no experimental history and were randomly divided with oral dabigatran doses into 1mg/kg, 5mg/kg,10mg/kg, 20mg/kg 40mg/kg,respectively and control group, 6 rabbits in each group.Blood was taken from the exposed femoral vein.To observe the changes of PT, APTT, TT and various coagulation factors activity(FⅡ、 FⅤ、 FⅧ、 FⅨ、 FⅩ、 FⅪ、 FⅫ) before and after taking medicine.Using the paired t test, Spearman rank correlation analysis and other statistical methods to analyze the experimental data, to understand the effect of dabigatran on the monitoring indicators, and study of its mechanism.Results:1、Comparedwith routine coagulation index(APTT、PT、TT)in each group were by paired t-test showed :APTT had no significant difference in the control group, 1mg / kg group. APTT in the 5mg/kg dose group prolonged with a statistical s ignificance(P < 0.05).PT has no difference in the control group, 1mg/kg group, 5mg/kg group, 10mg/kg group.PT prolonged in the 20mg/kg group, 40mg/kg group(P < 0.05). TT prolonged in the 1mg/kg group(P < 0.05). 2、Compare APTT, PT, TT of each group after medication with dabigatran dose by Spearman rank correlation analysis showed that APTT and TT showed a linear correlation with dabigatran dose(APTT:r=0.864, P < 0.01; TT : r = 0.797, P < 0.05). PT had no significant correlation with dabigatran dose(r=0.232, P=0.452). 3, all coagulation factor activity decline with dabigatran dose increases. coagulation factor activity has no differences in the control group and low dabigatran dose group(1mg/kg group). In the 5mg/kg group, all kinds of coagulation factors activity decreased(P < 0.05). The changes rate of coagulation factor activity suggest that coagulation factors(FⅧ、FⅨ、FⅪ)activity decreased more than coagulation factors(FII, FV, FX, FXII) activity. F XII activity decreased least than all coagulation factors.Conclusions:1, APTT, PT, TT can prolong at different level after taking dabigatran.2, APTT and TT showed a linear correlation with dabigatran dose. PT had no significant correlation with dabigatran dose.3, all kinds of coagulation factors activity decreased at different level after taking dabigatran. coagulation factors(FⅧ、FⅨ、FⅪ)activity decreased more than coagulation factors(FII, FV, FX, FXII) activity.
