Effect Of Qigui Granule On Expression Of IL-1and IFN-γ In Rats With Hypothyroidism

Objective: This study is through the observation of self-made prescription Qigui granule on thyroid function impairment effects of IL-1 and IFN-γ expression level in serum of rats and to observe the effect of Qigui granule on thyroid function decline, and to explore the mechanism of the treatment for hypothyroidism,and to explore the function of hypothalamus-pituitary-thyroid axis system,and clarify its mechanism of action.Methods: This experiment selected 50 Wistar rats, were randomly divided into six groups,10 in each group.That is blank group, model group, western medicine group and Chinese traditional medicine group which contain low dose group and high dose group of Qigui granule. To the saline lavage except the blank group, the rest of the group are 0.1%Propylthiouracil(PTU) saline solution, made hypothyroidism model.After 15 days, testing thyroid function and detecting IL-1 and IFN-γ levels, confirmed that the model is successful,after receiving the payment corresponding continuous treatment of western medicine, Qigui granule infusion of 8 weeks.After 8 weeks, unified collection and detection FT3 FT4, TSH,TG-Ab, TPO-Ab.Using the method of electrochemiluminescence.At the same time,detection IL-1 and IFN-γ levels by ELISA kits.Results: Hypothyroidism in rats after modeling after administration of drug treatment, the symptoms of hypothyroidism, the symptoms were improved in different degree.After 8weeks of treatment, TCM each dose group of FT3, FT4 were higher than the model group,there were significant difference( P <0.05);TCM each dose group of TSH were lower than the model group, there were significant difference( P <0.05); Compared with lower Chinese medicine dose group, high Chinese medicine dose group, the levels of FT3,FT4,TSH,there were very significant difference( P <0.01); Compared with western medicine group, high Chinese medicine dose group, the levels of FT3,FT4,TSH,there were no significant difference( P >0.05). TCM each dose group of IL-1, IFN-γ levels were lower than the model group, there were significant difference( P <0.05); Compared with western medicine group,the low Chinese medicine dose group, the levels of IL-1, IFN-γ levels,there were no significant difference( P >0.05);Compared with blank group, the high Chinese medicine dose group, the levels of IL-1, IFN-γ levels,there were no significant difference( P >0.05);Compared with western medicine group, the high Chinese medicine dose group,the levels of IL-1, IFN-γ levels,there were very significant difference( P <0.01). After administration, the serum levels of TG-Ab and TPO-Ab were not significantly improved,and there was no statistical significance( P > 0.05).Conclusion: Chinese medicine different dose groups of Qigui granule can significantly improve the general state of hypothyroid rats, improve the FT3, FT4 levels and reduce TSH levels. high Chinese medicine dose group of Qigui granule can significantly reduce IL-1,IFN-γ levels and improve clinical symptoms of hypothyroidism.
